Target Background

Function
Phospholipid scramblase involved in autophagy and cytoplasm to vacuole transport (Cvt) vesicle formation. Cycles between the preautophagosomal structure/phagophore assembly site (PAS) and the cytoplasmic vesicle pool and supplies membrane for the growing autophagosome. Lipid scramblase activity plays a key role in preautophagosomal structure/phagophore assembly by distributing the phospholipids that arrive through ATG2 from the cytoplasmic to the luminal leaflet of the bilayer, thereby driving autophagosomal membrane expansion. Required for mitophagy. Also involved in endoplasmic reticulum-specific autophagic process and is essential for the survival of cells subjected to severe ER stress. Recruits vesicle-tethering proteins TRS85 and YPT1 to the autophagosome formation site. Recruits also ATG23 and ATG8 to the PAS.

  2. This study identified Pho23 as a master transcriptional repressor for autophagy that regulates the frequency of autophagosome formation through its negative regulation of ATG9. PMID: 24881874
  3. PAS localization of Atg9 was normal in atg27(DeltaYSAV) cells. PMID: 25557545
  4. ATG9 trafficking to phagophore assembly site depends on ATG23 and ATG27 stochiometry. PMID: 25385507
  5. TRAPPIII(trs85) is responsible for vesicular transport from early endosomes to Golgi, facilitating Atg9 cycling in autophagy. PMID: 23986483
  6. Data show the PtdIns 4-kinase Pik1 is involved in Atg9 trafficking through the Golgi and is involved in both nonselective and selective types of autophagy, whereas the PtdIns4P 5-kinase Mss4 is involved in mitophagy but not nonselective autophagy. PMID: 22977244
  7. It was shown that that the vast majority of Atg9 existed on cytoplasmic mobile vesicles that were derived from the Golgi apparatus in a process involving Atg23 and Atg27. Only a few Atg9 vesicles were needed for a single round of forming autophagosomes. PMID: 22826123
  8. Data show that membrane that contains Atg9 is delivered to the autophagosome from the Golgi-endosomal system rather than from the ER or mitochondria. PMID: 20861302

  11. we demonstrate that Atg9 shuttles between the preautophagosomal structure and mitochondria PMID: 16874040
  12. Atg27 is required for Atg9 cycling and shuttles between the pre-autophagosomal structure, mitochondria, and the Golgi complex. PMID: 17135291
  13. model proposed for Atg9 cycling suggests pool of Atg11 mediates anterograde transport of Atg9 to the pre-autophagosomal structure/phagophore assembly site along the actin cytoskeleton PMID: 17329962
  14. Results support a model in which Atg9 multimerization facilitates membrane flow to the phagophore assembly site for phagophore formation. PMID: 18829864
  15. Atg9 can both promote lipid transport from the membrane origins to the phagophore assembly site, and also help assemble an intact phagophore membrane PMID: 19182520
