Target Background

Function
Serine/threonine-protein kinase. Needed for the initiation of DNA synthesis during mitosis as well as for synaptonemal complex formation and commitment to recombination during meiosis. Required for HTA1-HTB1 locus transcription repression.
Gene References into Functions
  1. main conclusion from our studies is that Cdc7 kinase is found at many locations in the genome, but is enriched at functional origins of replication PMID: 29134273
  2. Cdc7 is loosely bound to chromatin during G1. At the G1/S transition, Cdc7 binding to chromatin is increased and stabilized, preferentially at sites that may become replication origins. PMID: 28924058
  3. Data show that Dbf4-dependent kinase (DDK)-phosphoryled MCM DNA helicase complex subunit MCM2 preferentially interacts with DNA replication initiation factor CDC45 PMID: 26305950
  4. Study provides evidence that a key prerequisite for double-strand breaks formation-phosphorylation of Mer2 by Dbf4-dependent Cdc7 kinase, DDK-is linked to replication via association of the kinase with replisome components. PMID: 25126790
  5. *Phosphorylation of Mcm2-7 double hexamers by Dbf4-dependent kinase promotes efficient recruitment of overexpressed firing factors from S-phase extracts. PMID: 24566989
  6. Kinetochores coordinate pericentromeric cohesion and early DNA replication by Cdc7-Dbf4 kinase recruitment. PMID: 23746350
  7. Dbf4 and Cdc7 proteins promote DNA replication through interactions with distinct Mcm2-7 protein subunits. PMID: 23549044
  8. Inhibition of Cdc7 by the DNA damage response prevents Eco1 destruction, allowing establishment of cohesion after S phase. PMID: 23314252
  9. Cdc7-Dbf4 promotes NDT80 transcription by relieving repression mediated by a complex of Sum1, Rfm1, and a histone deacetylase, Hst1. PMID: 22106412
  10. Studies identify distinct roles for DDK and S-CDK during helicase activation and support a model in which the leading strand DNA polymerase is recruited prior to origin DNA unwinding and RNA primer synthesis. PMID: 21729781
  11. multiple phosphorylation sites within Rec8 as well as two different kinases, casein kinase 1delta/epsilon (CK1delta/epsilon) and Dbf4-dependent Cdc7 kinase (DDK), are required for Rec8 cleavage and meiosis I nuclear division PMID: 20230747
  12. regions on budding yeast Dbf4 required for binding and activating Cdc7 kinase PMID: 19822727
  13. the amino terminal serine/threonine-rich domain (NSD) of Mcm4 has both inhibitory and facilitating roles in DNA replication control and the sole essential function of DDK is to relieve an inhibitory activity residing within the NSD PMID: 20054399
  14. An extensive genetic analysis of the interactions between CDC7 and REV3, RAD30, RAD5, or POL30 in response to DNA damage was done to determine its role in the RAD6 pathway. PMID: 15342501
  15. Cdc7-Dbf4 protein kinase (DDK) promotes assembly of a stable Cdc45-MCM complex exclusively on chromatin in S phase. PMID: 17018296
  16. Cdc7-Dbf4 phosphorylation of the MCM complex normally results in a single, highly active conformation of Mcm5, whereas the mcm5-bob1 mutation produces a number of conformations, only one of which is permissive for origin activation. PMID: 17724082
  17. Coordination between premeiotic S and DSB formation may be achieved by using CDK-S and DDK to initiate both processes. PMID: 18245450
  18. Cdc7, in concert with CDK, regulates Spo11 loading to DSB sites via Mer2 phosphorylation. PMID: 18245451
  19. Cdc7 allows expression of NDT80, is necessary for recruitment of monopolin to sister kinetochores, and it is necessary for the reductional segregation occurring at meiosis I. PMID: 18768747
  20. Dbf4-dependent Cdc7 kinase (DDK) regulates monopolin localization together with the polo-kinase Cdc5 bound to Spo13, probably through phosphorylation of the monopolin subunit Lrs4 PMID: 19013276
  21. incorporation of Mcm2-7 into the prereplicative complex increased the level and changes the specificity of Dbf4-dependent kinase phosphorylation of this complex PMID: 19270162
  22. Cdc7p-Dbf4p prevents inappropriate exit from mitosis by inhibiting Polo kinase and functions in the spindle position checkpoint. PMID: 19478884
  23. Dbf4-Cdc7 phosphorylation of Mcm2 is required for cell growth PMID: 19692334
