Target Background

Function
ATP-dependent RNA helicase involved in mRNA turnover, and more specifically in mRNA decapping by activating the decapping enzyme DCP1. Is involved in G1/S DNA-damage checkpoint recovery, probably through the regulation of the translational status of a subset of mRNAs. May also have a role in translation and mRNA nuclear export. Required for sporulation. Blocks autophagy in nutrient-rich conditions by, at least partly, binding and repressing the expression of a set of ATG genes, including ATG3, ATG7, ATG8, ATG19, ATG20, ATG22 and SNX4/ATG24.
Gene References into Functions
  1. All Dhh1-dependent mRNAs, viral and cellular, share key common features. First, they contain long and highly structured CDSs, including a region located around nucleotide 70 after the translation initiation site; second, they are directly bound by Dhh1 with a specific binding distribution; and third, complementary experimental approaches suggest that they are activated by Dhh1 at the translation initiation step PMID: 27821408
  2. Here, the authors show that in budding yeast, mutations in the DEAD-box ATPase Dhh1 that prevent ATP hydrolysis, or that affect the interaction between Dhh1 and Not1, the central scaffold of the CCR4-NOT complex and an activator of the Dhh1 ATPase, prevent processing bodies disassembly in vivo. PMID: 27692063
  3. findings show that the mutations in ATPase A and B motif (K96R, D195A) affected the growth at non-permissive temperature, the mating processes including the Ste12 expression, and the P-body accumulation PMID: 28455591
  4. These results suggest that after localization to the bud tip, a portion of the localized ASH1 mRNA becomes translationally inactive because of binding of Dhh1 and Puf6 to the 5'- and 3'-UTRs of ASH1 mRNA. PMID: 28450395
  5. Data indicate that RNA helicase RCK family member Dhh1 and the mRNA decay pathway coordinately repress the autophagy gene expression. PMID: 26098573
  6. Dhh1 activates decapping in response to impaired ribosome elongation. PMID: 22719226
  7. Pull-down experiments reveal that the intracellular binding partners of Dhh1 change as cells undergo the diauxic growth shift. PMID: 21712243
  8. Results place Dhh1 at the interface of translation and decay controlling whether an mRNA is translated, stored, or decayed. PMID: 21844211
  9. Pbp1 and Dhh1 over-expression inhibits cell growth, and for Dhh1, leads to the accumulation of stress granules PMID: 20368989
  10. DHH1 has a role in G1/S DNA-damage checkpoint recovery PMID: 15166134
  11. The Dhh1p is as functioning as translational repressors and facilitators of P body formation. PMID: 16179257
  12. Caf20 and Dhh1 control filamentation by regulating STE12 translation. PMID: 17041186
  13. We propose that Dhh1 and Caf20 regulate the Ste12 protein expression and the Ste12 protein level is associated with P-body formation during mating. PMID: 18182159
