Target Background

Function
Ubiquitin-binding protein involved in protein ubiquitination, sorting and degradation. Acts as a ubiquitinated substrate-recruiting adapter for chaperone ATPase CDC48 by binding mono- or polyubiquitin chains. Depending on the context, promotes or prevents proteasomal degradation of ubiquitinated proteins. Involved in the ubiquitin fusion degradation (UFD) pathway by promoting the degradation of ubiquitinated proteins. Involved in the mitochondria-associated degradation pathway (MAD) by promoting the degradation of several ubiquitinated membrane proteins. By competing with UFD2 to bind CDC48, prevents the multi-ubiquitination and subsequent degradation of UFD2-dependent substrates. Required for ribophagy, a process which relocalizes ribosomal particles into the vacuole for degradation in response to starvation. Involved in the ubiquitin-mediated sorting of membrane proteins into multivesicular bodies (MVBs). In addition, plays an essential role in maintaining cellular ubiquitin levels. May affect indirectly the degradation of ubiquitinylated proteins by regulating cellular ubiquitin levels.
Gene References into Functions
  1. Small-angle X-ray scattering combined with molecular docking and biochemical analysis yield the solution structure of the Doa1/PFU:Hse1/SH3 complex and suggest that hydrogen bonding is a major determinant in the interaction of Doa1/PFU with Hse1/SH3. PMID: 24607902
  2. A structural comparison of Doa1 with Ufd2 revealed that they share a similar ARM-like repeat, supporting a model in which Doa1 and Ufd2 compete for Cdc48 binding and may dictate the fate of ubiquitinated proteins in the proteasome pathway PMID: 21063153
  3. The data suggest that Doa1 is the major source of ubiquitin for the DNA damage response and that Doa1 also plays an additional essential and more specific role in the monoubiquitination of histone H2B. PMID: 16705165
  4. DOA1/UFD3 plays a role in sorting ubiquitinated membrane proteins into multivesicular bodies PMID: 18508771
