Target Background

Function
Component of the cytosolic iron-sulfur (Fe-S) protein assembly (CIA) machinery required for the maturation of extramitochondrial Fe-S proteins. Part of an electron transfer chain functioning in an early step of cytosolic Fe-S biogenesis, facilitating the de novo assembly of a [4Fe-4S] cluster on the scaffold complex CFD1-NBP35. Electrons are transferred to DRE2 from NADPH via the FAD- and FMN-containing protein TAH18. TAH18-DRE2 are also required for the assembly of the diferric tyrosyl radical cofactor of ribonucleotide reductase (RNR), probably by providing electrons for reduction during radical cofactor maturation in the catalytic small subunit RNR2. Has anti-apoptotic effects in the cell. Involved in negative control of H(2)O(2)-induced cell death.
Gene References into Functions
  1. the pathways for RNR and Fe-S cluster biogenesis bifurcate after the Dre2-Tah18 step. PMID: 28515324
  2. The conserved protein Dre2 uses essential [2Fe-2S] and [4Fe-4S] clusters for its function in cytosolic iron-sulfur protein assembly PMID: 27166425
  3. each of the Cys Fe-S cluster ligands with the exception of C252 is essential, indicating that both Dre2 clusters are needed for cell viability. PMID: 27672211
  4. Study show that Dre2 contains an N-terminal domain whose structure in solution consists of a stable well-structured monomer with an overall typical S-adenosylmethionine methyltransferase fold lacking two alpha-helices and a beta-strand. PMID: 22487307
  5. Interaction between the Tah18 and Dre2 C-terminus is essential for viability. PMID: 21902732
  6. Data show that Dre2 is an essential conserved Fe/S cluster protein implicated in extramitochondrial Fe/S cluster assembly, although partially localized to the mitochondrial intermembrane space. PMID: 18625724
  7. These results are in favour of an oxidative stress-induced cell death in yeast that involves mitochondria and is controlled by the newly identified Dre2-Tah18 complex. PMID: 19194512
