Target Background

Function
Catalytic subunit of the RNA polymerase II elongator complex, which is a component of the RNA polymerase II (Pol II) holoenzyme and is involved in transcriptional elongation. Association with elongating RNAPII requires a hyperphosphorylated state of the RNAPII C-terminal domain (CTD). The elongator complex is required for multiple tRNA modifications, including mcm5U (5-methoxycarbonylmethyl uridine), mcm5s2U (5-methoxycarbonylmethyl-2-thiouridine), and ncm5U (5-carbamoylmethyl uridine). ELP3 acts as a tRNA uridine(34) acetyltransferase, which mediates formation of carboxymethyluridine in the wobble base at position 34 in tRNAs. Independently, ELP3 may be involved in polarized exocytosis.
Gene References into Functions
  1. Results conclude that yeast and plant Elongator (elp3/ELO3) share tRNA modification roles and propose that this function might be conserved in Elongator from all eukaryotic kingdoms of life. PMID: 20398216
  2. the Elp3 FeS cluster is essential for normal Elongator function in vivo primarily as a structural, rather than catalytic, domain. PMID: 18986986
  3. report an interaction between the Elongator histone acetyltransferase and the proliferating cell nuclear antigen. Cells lacking Elp3 and are sensitive to the DNA replication inhibitor hydroxyurea and the damaging agent methyl methanesulfonate. PMID: 19834596
