Target Background

Function
Component of the ribosome, a large ribonucleoprotein complex responsible for the synthesis of proteins in the cell. The small ribosomal subunit (SSU) binds messenger RNAs (mRNAs) and translates the encoded message by selecting cognate aminoacyl-transfer RNA (tRNA) molecules. The large subunit (LSU) contains the ribosomal catalytic site termed the peptidyl transferase center (PTC), which catalyzes the formation of peptide bonds, thereby polymerizing the amino acids delivered by tRNAs into a polypeptide chain. The nascent polypeptides leave the ribosome through a tunnel in the LSU and interact with protein factors that function in enzymatic processing, targeting, and the membrane insertion of nascent chains at the exit of the ribosomal tunnel. Located at the head of the 40S ribosomal subunit in the vicinity of the mRNA exit channel, RACK1 serves as a scaffold protein that can recruit other proteins to the ribosome. Involved in induction of the ribosome quality control (RQC) pathway; a pathway that degrades nascent peptide chains during problematic translation. Involved in the negative regulation of translation of a specific subset of proteins.
Gene References into Functions
  1. Data provide evidence that Asc1 seems to connect signal transduction not only with the translational machinery, but also with fundamental nuclear processes. PMID: 28982715
  2. These indicate that ribosome-associated Asc1/RACK1 facilitates endonucleolytic cleavage of nonstop mRNA by stalled ribosomes and represses the levels of aberrant products even in the absence of Dom34. We propose that Asc1/RACK1 acts as a fail-safe in quality control for nonstop mRNA. PMID: 27312062
  3. identify Asc1p (aka RACK1) and Rps3p, both stable 40S subunit components, as additional 18S nonfunctional 18S rRNA decay factors PMID: 28956756
  4. It is the paper where ASC1 gene is identified and cloned and named. The effect of ASC1 deletion is also studied. PMID: 9504906
  5. Translation fidelity depends upon Asc1-mediated quality control. PMID: 25792604
  6. These results indicate the intertwined structure of a relatively small and stable protein core and the remaining regions of different levels of flexibility. PMID: 24591271
  7. Genetic interactions between dys1-1 mutants and mutant PKC1 and ASC1, implicate a role for HYP2 (eIF5A homolog) and Asc1 in the maintenance of cell integrity at the translational level. PMID: 23573236
  8. Evidence of the translational regulation of such transcription factors suggests that ribosomal Asc1p is involved in signal transduction pathways and controls the biosynthesis of the respective final transcriptional regulators. PMID: 23071099
  9. The structure reveals an atypical mode of dimerization where monomers intertwine on blade 4, thus exposing a novel surface of the protein to potential interacting partners. PMID: 21704636
  10. Asc1 functions as a G-protein beta subunit coupled to glucose responsiveness in yeast PMID: 17591772
  11. RACK1 functions from the ribosome, implying a physical link between the eukaryotic ribosome and cell signaling pathways in vivo. PMID: 19114558
