Target Background

Function
Cleaves the gamma-glutamyl bond of periplasmic glutathione (gamma-Glu-Cys-Gly), glutathione conjugates, and other gamma-glutamyl compounds. The metabolism of glutathione releases free glutamate and the dipeptide cysteinyl-glycine, which is hydrolyzed to cysteine and glycine by dipeptidases; it may function in amino acid uptake/salvage, or possibly in peptidoglycan linkage. Catalyzes the hydrolysis and transpeptidation of many gamma-glutamyl compounds (including some D-gamma-glutamyl substrates), with a preference for basic and aromatic amino acids as acceptors. The KM values for gamma-glutamyl acceptors are so high that it has been proposed transpeptidation is not the physiological role in E.coli.
Gene References into Functions
  1. beta-N-(gamma-L(+)-glutamyl)phenylhydrazine was prepared from L-glutamine and phenylhydrazine through a transpeptidation reaction of Escherichia coli gamma-glutamyltranspeptidase (although phenylhydrazine has been reported to be an inhibitor of the enzyme). PMID: 22791224
  2. In contrast to the human GGT1, the E. coli enzyme was a nonselective enzyme that accommodated substrates without specifically recognizing the C-terminal carboxy group of the Cys-Gly moiety PMID: 17260973
  3. Data describe the crystal structures of Escherichia coli gamma-glutamyltranspeptidase in complex with azaserine and acivicin, and suggest a novel method for inhibition by glutamine antagonists. PMID: 18555071
  4. the Ser463 residue contributes to the enzymatic and autoprocessing activities of Escherichia coli gamma-glutamyltranspeptidase PMID: 19788059
