Target Background

Function
Component of the glutathione system which performs several activities such as glutathione-dependent oxidoreductase, glutathione peroxidase and glutathione S-transferase (GST) activity. The disulfide bond functions as an electron carrier in the glutathione-dependent synthesis of deoxyribonucleotides by the enzyme ribonucleotide reductase. In addition, it is also involved in reducing cytosolic protein- and non-protein-disulfides in a coupled system with glutathione reductase. Required for resistance to reactive oxygen species (ROS) by directly reducing hydroperoxides and for the detoxification of ROS-mediated damage. GRX2 is more active as an oxidoreductase than GRX1. Responsible for the S-glutathionylation of DHBP synthase.
Gene References into Functions
  1. Data show that the gatekeeper function of glutaredoxin Grx2 is dependent on its concentration in the mitochondrial intermembrane space (IMS). PMID: 25392302
  2. specific functions of two interacting redoxins: dually localized glutaredoxin 2 (Grx2p) and mitochondrial peroxiredoxin 1 (Prx1p) PMID: 21385868
  3. Results suggest that although Trx is the reductant involved in the reduction of peroxides by 2-Cys-peroxiredoxin (Prx), glutaredoxin 2 might be the natural resolving partner of 1-Cys Prx through a monothiolic mechanism. PMID: 20059400
  4. Mutations on key basic residues drastically affect the cellular fate of the protein, showing that evolutionary diversification of Grx2 structural and functional properties are strictly dependent on the sequence of the targeting signal peptide. PMID: 20036764
  5. a plethora of Grx2 subcellular localizations could spread its antioxidant functions all over the cell, but one single Ala to Gly mutation converts Grx2 into a typical protein of the mitochondrial matrix PMID: 16606613
  6. Grx2 is involved with the apoptotic death induced by cadmium, a form of cellular suicide that might lead of removal of mutated cells. PMID: 17996374
  7. glutaredoxin 2 and cytosolic thioredoxins have roles in cysteinyl-based redox modification of the 20S proteasome PMID: 18435761
