Target Background

Function
Catalyzes the reversible interconversion of serine and glycine with tetrahydrofolate (THF) serving as the one-carbon carrier. This reaction serves as the major source of one-carbon groups required for the biosynthesis of purines, thymidylate, methionine, and other important biomolecules. Also exhibits THF-independent aldolase activity toward beta-hydroxyamino acids, producing glycine and aldehydes, via a retro-aldol mechanism. Thus, is able to catalyze the cleavage of allothreonine and 3-phenylserine. Also catalyzes the irreversible conversion of 5,10-methenyltetrahydrofolate to 5-formyltetrahydrofolate.
Gene References into Functions
  1. A bioinformatic study shows that Tyr55 (at the active site) and Arg235 are strongly conserved within the serine hydroxymethyltransferase family and are involved in a cation-pi interaction in the large majority of available SHMT structures. PMID: 19883126
  2. activation of glyA by CsgD requires the participation of the regulatory proteins, MetR and PurR PMID: 15644921
  3. Results describe the mechanism of addition of pyridoxal 5'-phosphate to Escherichia coli apo-serine hydroxymethyltransferase. PMID: 17341210
