Target Background

Function
S-adenosyl-L-methionine-dependent protein-arginine N-methyltransferase that catalyzes both the mono- and asymmetric (type I) dimethylation of the guanidino nitrogens of arginine residues in a variety of RNA-binding proteins such as heterogeneous nuclear ribonucleoproteins (hnRNPs) and small nuclear ribonucleoproteins (snRNPs). Methylates NAB2, NPL3, HRP1 and YRA1, shuttling hnRNPs involved in mRNA processing and export, facilitating their export out of the nucleus. Methylation of NPL3 weakens its interaction with THO2, a component of the TREX (transcription/export) complex important for transcriptional elongation and recruitment of mRNA export factors. Methylates the hnRNP HRB1, but does not influence its subcellular location. Methylates the nucleolar proteins GAR1, NOP1 and NSR1. Methylates the snRNP SNP1 and modulates the cotranscriptional recruitment of splicing factors. Dimethylates free histone H4 (HHF1/HHF2) at 'Arg-4' (H4R3me2a) and plays a role in preservation and establishment of silent chromatin domains. Mono- and dimethylates ribosomal protein S2 (RPS2) at 'Arg-11'. Methylates the catalytic subunit of the SWI/SNF chromatin-remodeling complex SNF2.
Gene References into Functions
  1. Data suggest that Hmt1-based arginine methylation is required for Scd6 localization and function. PMID: 27776129
  2. Arginine methylation is a widespread modification, with Hmt1 being the major arginine methyltransferase in S. cerevisiae. PMID: 26572822
  3. the methyltransferase activity of Hmt1 on histone H3R2 requires reciprocal contributions from two Hmt1 molecules, suggesting an intermolecular trans-complementary mechanism by which Hmt1 dimer methylates its substrates PMID: 25715670
  4. Carried out a proteomic profiling experiment to comprehensively identify the physical interactors of Hmt1, the budding yeast homolog for human PRMT1. Snf2 was experimentally confirmed as a robust substrate of Hmt1 in vitro. PMID: 22997150
  5. Interactions between HMT1 and genes encoding components of the histone deacetylase complex Rpd3L, are reported. PMID: 22953000
  6. These results are suggestive of a role for Rmt1 in modifying the function of Rps2 in a manner distinct from that occurring in S. pombe and mammalian cells. PMID: 20035717
  7. Loss of Hmt1's catalytic activity results in dimethylated Arg-3 histone H4 occupancy across silent chromatin regions. PMID: 17158743
  8. Results reveal a functional connection between histone acetylation, methylation, and two of the responsible enzymes, Gcn5p and Hmt1p. PMID: 19358899
