Target Background

Function
Catalytic component of the TFIIK complex (KIN28-CCL1 dimer) which is the protein kinase component of transcription factor IIH (TFIIH) and phosphorylates the C-terminal domain of RNA polymerase II during transition from transcription to elongation after preinitiation complex (PIC) formation, thereby positively regulating transcription. TFIIH (or factor B) is essential for both basal and activated transcription, and is involved in nucleotide excision repair (NER) of damaged DNA. TFIIH has DNA-dependent ATPase activity and is essential for polymerase II transcription in vitro. Essential for cell proliferation.
Gene References into Functions
  1. Inactivating Kin28 impacts promoter release to differing degrees and reveals a "checkpoint" during the transition to productive elongation, resulting in widespread mRNA stabilization. PMID: 27477907
  2. Kin28 has a role in the association of Mediator with promoters. PMID: 24704787
  3. These results identify an Spt5 phosphorylated C-terminal repeats-independent pathway of Paf1C recruitment requiring Kin28, Bur1, and the Spt5 phosphorylated C-terminal repeats-interaction domain of Cdc73. PMID: 22796944
  4. These results suggest that the overexpression of Kin28 and Pog1 enhances the protein refolding and degradation pathways in rsp5(A401E) cells. PMID: 17986087
  5. Results implicate the TFIIH kinase in placing bivalent Ser5 and Ser7 marks early in gene transcription. PMID: 19450536
  6. Study points strongly toward a minor role of TFIIH kinase in Pol II transcription, and a more significant role in mRNA capping in budding yeast. PMID: 19666497
