Target Background

Function
Required for branched chain fatty acid synthesis. Catalyzes the reduction of the 3-ketoacyl-CoA intermediate that is formed in each cycle of fatty acid elongation. Very long-chain fatty acids (VLCFAs) serve as precursors for ceramide and sphingolipids. Probably involved in hormone production as it metabolizes 4-androstendione (androst-4-ene-3,17-dione) into testosterone and estrone into estradiol (17beta-estradiol) in vitro, but the physiological steroid substrate is unknown.
Gene References into Functions
  1. LET-767 modifies a sterol hormone that is required both for embryogenesis and for later stages of development. PMID: 12905072
  2. High conservation of the active site and amino acids responsible for enzymatic activity and substrate selectivity strongly suggests that LET-767 shares a common ancestor with human 17beta-HSD3 and 12. PMID: 17951538
  3. LET-767 is a major 3-ketoacyl-CoA reductase in C. elegans required for the bulk production of monomethyl branched and long chain fatty acids, and the developmental arrest in let-767 (RNAi) worms is caused by the deficiency of the former PMID: 18390550
Protein Families
Short-chain dehydrogenases/reductases (SDR) family, 17-beta-HSD 3 subfamily
Tissue Specificity
Expressed in the gut of larva and adult.
