Target Background

Function
Central component of the spindle assembly checkpoint which is a feedback control that prevents cells with incompletely assembled spindles from leaving mitosis. Thought to be recruited to unattached kinetochores by MAD1. During checkpoint activity, MAD2 is relayed from the MAD1-MAD2 complex to the mitotic checkpoint complex (MCC). MCC and presumably the MAD2-CDC20 subcomplex inhibit the ubiquitin ligase activity of the anaphase promoting complex/cyclosome (APC/C) by preventing its activation by CDC20.
Gene References into Functions
  1. Mad2 recruitment to the kinetochore and it's role in the spindle assembly checkpoint PMID: 27280788
  2. the very same dimerization interface of Mad2 is required for two different, yet fundamental, consecutive steps of SAC signaling: at the kinetochores for priming Cdc20/Mad2 binding, away from kinetochores for stabilizing this complex PMID: 23000150
  3. Mad2 is an important meiotic cell cycle regulator that ensures the timely degradation of anaphase-promoting complex substrates and the proper orchestration of the meiotic divisions PMID: 21697504
  4. Mad2 prolongs DNA damage checkpoint arrest caused by a double-strand break via a centromere-dependent mechanism. PMID: 20096585
  5. Data demonstrate that Mad2p localization at nuclear pore complexes is sensitive to nuclear levels of Ran and suggest that release of Mad2p from NPCs is closely linked with spindle assembly checkpoint activation in yeast. PMID: 15701789
  6. Our biochemical and genetic evidence shows that the interaction of open and closed Mad2 is essential for the spindle assembly checkpoint and is conserved in evolution. PMID: 16818718
  7. results surpass previously formulated objections to the Mad2-template model and predict that the release of Mad2 from Cdc20 is an energy-driven process PMID: 19143472
