Target Background

Function
Component of the ESCRT-I complex, a regulator of vesicular trafficking process. Binds to ubiquitinated cargo proteins and is required for the sorting of endocytic ubiquitinated cargos into multivesicular bodies (MVBs). Appears to be involved in cargo sorting and release of the ESCRT-I complex from the MVBs.
Gene References into Functions
  1. Mvb12 may function in regulating the interactions of ESCRT-I with cargo and other proteins of the ESCRT machinery to efficiently coordinate cargo sorting and release of ESCRT-I from the multivesicular body vesicles. PMID: 17135292
  2. Mvb12 stabilizes ESCRT-I in an oligomeric, inactive state in the cytosol to ensure that the ordered recruitment and assembly of ESCRT-I and -II is spatially and temporally restricted to the surface of the endosome after activation of the MVB reaction. PMID: 17145965
  3. These data support a model in which Mvb12 impacts the selection of multivesicular body cargoes by modulating the cargo recognition capabilities of ESCRT-I. PMID: 17151358
Subcellular Location
Cytoplasm. Endosome. Late endosome membrane; Peripheral membrane protein.
