Target Background

Function
Seems to play a role in optimizing the host cell environment for viral replication without causing cell death by apoptosis. Enhances virus infectivity and pathogenicity. Probably involved in viral immune evasion mechanisms.; In infected CD4(+) T-lymphocytes, down-regulates cell surface expression of CD4, CD28, CD3, and MHC-I or MHC-II molecules.; Interferes with TCR signaling from the cell membrane. Interacts with CD247/TCRZ (TCR zeta chain) and exert potent down-regulation of cell surface TCR/CD3 complexes.
Gene References into Functions
  1. the structure of SIVmac239 Nef bound to the ExxxLM motif of another Nef molecule at 2.5 A resolution, is reported. PMID: 28874665
  2. The convergent evolution of nef sequences in the central nervous system of Rhesus Macaques suggests a significant role for nef in establishing neurotropic strains. PMID: 26041280
  3. Nef is not involved in protecting SIV-infected macrophages from CD8+T-cell killing. PMID: 26269172
  4. Simian immunodeficiency virus Nef-induced inhibition of TfR endocytosis leads to the reduction of Transferrin uptake and intracellular iron concentration and is accompanied by attenuated lentiviral replication in macrophages. PMID: 24383984
  5. Data suggest that Simian immunodeficiency virus Nef protein and rhesus macaque tetherin/BST2 (bone marrow stromal antigen 2) undergo physical interaction leading to removal of tetherin from plasma membrane by clathrin-mediated endocytosis. PMID: 23853598
  6. study identifed three amino-acids in the C-terminal domain of Nef that are critical specifically for its ability to antagonize Tetherin PMID: 21625568
  7. We explored T lymphocyte recognition of a dominant Nef epitope of SIVmac251 in infected Mamu-A*02(+) rhesus monkeys PMID: 21841125
  8. we report here that mutation of the diacidic motif in the PBj-Nef protein abolishes disease progression in pig-tailed macaques despite efficient replication. PMID: 21366921
  9. These results indicate that simian immunodeficiency virus infection is a significant contributor to primary dyslipidemia, likely through the ability of Nef to suppress ABCA1-dependent reverse cholesterol transport. PMID: 20617930
  10. nef serves to limit the virus-specific CD8 cellular response of the host and that the ability to downregulate MHC class I contributes importantly to the totality of nef function. PMID: 15542684
  11. it was demonstrated that protein kinase CK2 (CK2) specifically associates with and phosphorylates SIV/17E-Fr Nef PMID: 16448682
  12. both the Nef functions that were disrupted by the delta64-67 mutation and the activities that remained intact contribute to viral pathogenicity PMID: 16611907
  13. Discovery of complex vascular lesions in SHIV-nef but not SIV-infected animals, the presence of Nef in vascular cells of patients with HIV-related pulmonary hypertension suggests Nef plays a key role in development of severe pulmonary arterial disease. PMID: 16728715
  14. SIV-1 Nef regulates intracellular ion concentrations indirectly. PMID: 18489774
  15. T cell receptor zeta forms a 1:1 complex with Nef and remains unfolded in the complex PMID: 19012413
  16. nef alleles from all SIVsmm lineages down-modulated CD4, MHC-I, CD28 and CD3 and up-regulated the invariant chain (Ii) associated with immature MHC-II molecules in human-derived cells PMID: 19358735
  17. findings show that SIV Nef overcomes restriction by rhesus macaque & sooty mangabey tetherin, but not human tetherin; specificity of SIV Nef for rhesus tetherin mapped to a four amino acid sequence in the cytoplasmic domain PMID: 19436700
