Target Background

Function

Rep helicase is a single-stranded DNA-dependent ATPase involved in DNA replication; it can initiate unwinding at a nick in the DNA. It binds to the single-stranded DNA and acts in a progressive fashion along the DNA in the 3' to 5' direction.

After replication blockage by an inverted rrn, Rep in conjunction with DinG or UvrD removes RNA polymerase, a task that is fulfilled in its absence by the SOS-induced DinG and UvrD helicases. PMID: 19851282
Rep helicase activity is regulated by the rotational conformational state of its 2B subdomain; formation of a Rep dimer may relieve autoinhibition by altering the 2B subdomain orientation. PMID: 16009938
