Target Background

Function
Essential for cell cycle arrest at the G1 and G2 stages following DNA damage by X-, and UV-irradiation, or inactivation of DNA ligase. Plays a role in the response to DNA damage. Interaction with rad4 via its phosphorylation sites in the N-terminus couples the DNA checkpoint apparatus to chromatin via interaction of its C-terminal BRCT domains with epigenetic modifications on histones H4 and H2A, respectively, in the G1/S phase of the cell cycle, and facilitates recruitment of the checkpoint kinase chk1.
Gene References into Functions
  1. Data show that polar interactions of Crb2 serine-548 and lysine-619 with the phosphate group of phospho-H2A (gamma-H2A) are critical for Crb2 IRIF formation and checkpoint function. PMID: 20679485
  2. Findings argue that binding of the Crb2 BRCT repeats to pH2AX is critical for checkpoint activity and provide new insight into the mechanisms of chromatin-mediated genome stability. PMID: 20679488
  3. When the Crb2 mutants experience damaged replication forks upon inhibition of the religation activity of topoisomerase I, the Chk1 DNA damage pathway induces sustained activation of the spindle checkpoint PMID: 16107732
  4. Full Crb2 activity requires phosphorylation of threonine-215 by Cdc2. PMID: 16314498
  5. Histone modifications cooperate in the same Crb2 recruitment pathway, which also requires the Tudor and BRCT motifs in Crb2. PMID: 16778077
  6. Structural and functional analysis of the crb2 domain reveals distinct roles in checkpoint signaling and DNA damage repair. PMID: 18676809
