Target Background

Function
Catalytic component of the COMPASS (Set1C) complex that specifically mono-, di- and trimethylates histone H3 to form H3K4me1/2/3, which subsequently plays a role in telomere length maintenance and transcription elongation regulation.
Gene References into Functions
  1. Set1 lacking RNA recognition motif 2 (RRM2) showed reduced in vivo cross-linking to RNA and reduced chromatin occupancy. In addition, levels of H3K4 trimethylation were decreased, whereas levels of dimethylation were increased PMID: 28483910
  2. Set1 and Set5 promote gene silencing at telomeric regions. Double mutants Set1 and Set5 display defects in telomere stability. PMID: 27911222
  3. Careful tuning of Set1 levels by regulated degradation is critical for the establishment and maintenance of proper H3K4 methylation patterns. PMID: 24613354
  4. The study uncovers novel pathways in which the methyltransferase Set5 participates and, more importantly, reveals a partnership between Set1 and Set5 in transcriptional repression near repetitive DNA elements in budding yeast. PMID: 24442241
  5. A number of aromatic amino acids in the SET domain of Set1 contribute to the formation of an active site that facilitates substrate binding and dictates product specificity. PMID: 23469257
  6. H3K4 methyltransferase Set1 is involved in maintenance of ergosterol homeostasis and resistance to Brefeldin A. PMID: 23382196
  7. The n-SET domain of Set1 is a predictor of an H2B ubiquitylation-sensing mechanism that leads to downstream H3K4 methylation. PMID: 23453808
  8. These results indicate that in S. cerevisiae, the non-redundant role of H3K4 methylation by Set1 is repression, achieved through promotion of 3'-end antisense transcription to achieve specific rather than global effects through two distinct mechanisms. PMID: 23028359
  9. Set1 is required for full repression of genes involved in ribosomal biosynthesis during diamide stress. PMID: 22912562
  10. Set1 is required for meiotic S-phase onset, double-strand break formation and middle gene expression. PMID: 15071505
  11. The catalytic domain of Set1 integrates opposing inputs from the the RNA-recognition motif and autoinhibitory domains to link properly H3K4 methylation to the transcript elongation process. PMID: 15775977
  12. Set1 trimethyl-defective mutants can rescue a set1Delta slow growth defect but are defective om telomere and rDNA silencing. PMID: 15964832
  13. Absence of the Set1 histone methyltransferase leads to a delay of premeiotic S phase by separate mechanisms. PMID: 16254243
  14. determined which members of COMPASS are required for silencing at the ribosomal DNA locus (rDNA), a telomere, and the silent mating loci (HM) PMID: 16582434
  15. Set1 RNA recognition motif (RRMs) bind RNA, but Set1 RNA-binding activity is not linked to H3K4 methylation. PMID: 16787775
  16. This study reports that Set1C subunits stimulate Set1 catalytic activity all along active genes. PMID: 16921172
  17. In this review, we discuss the regulation of H3K4 methylation and try to link Set1 activity with the multiple phenotypes displayed by cells lacking Set1. We also suggest that Set1 may have multiple targets. PMID: 16936826
  18. Histone H2B monoubiquitination by Rad6/Bre1 is required for the trimethylation of both histone H3K4 and H3K79 by set1 methyltransferase. PMID: 19667127
  19. These results support the idea that SET1 mRNA binding is dependent on translation and that SET1 mRNA-associated complex assembles on nascent Set1 in a cotranslational manner. PMID: 19713935
