Target Background

Function
Scaffold protein that most likely acts in the postsynaptic density (PSD) of excitatory synapses which orchestrates synapse formation and maintenance at neuromuscular junctions. Associates with and trafficks the L-type calcium channel egl-19 to the cell surface of body wall muscles to ensure the function of the calcium channel and therefore maintain the Ca(2+) current density. The maintenance of Ca(2+) also allows for the downstream regulation of Ca(2+)-induced expression of genes such as gem-4. Plays a role in the regulation of the defecation cycle, and this may be in association with the inositol trisphosphate (IP3) receptor itr-1, which in turn mediates periodic calcium release and muscle contractions. Required for normal fertility and pharyngeal pumping.
Gene References into Functions
  1. Here the authors show that the Caenorhabditis elegans SHN-1/Shank binds L-type calcium channels and that increased and decreased shn-1 gene dosage alter L-channel current and activity-induced expression of a CRH-1/CREB transcriptional target (gem-4 Copine). PMID: 28477407
  2. Shank ANK repeat-domain along with PDZ may play a crucial role in regulating calcium signaling with inositol triphosphate receptor PMID: 21191812
  3. SHN-1 protein is localized in various tissues including neurons, pharynx and intestine. Suppression of SHN-1 did not cause lethality or developmental abnormality, but resulted in altered defecation rhythm in a strain with mutated IP(3)receptor. PMID: 15013747
Subcellular Location
Cell projection, pseudopodium. Cytoplasmic vesicle. Cell junction, synapse, postsynaptic density.
Protein Families
SHANK family
Tissue Specificity
Expressed in the pharynx, pharyngeal-intestinal valve, intestine, rectal epithelial cells, tail neurons, nerve cord and sperm.
