Target Background

Function
Ligand for the EGF receptor (Gurken). Involved in a number of unrelated developmental choices, for example, dorsal-ventral axis formation, glial migration, sensory organ determination, and muscle development. It is required for photoreceptor determination.
Gene References into Functions
  1. We provide evidence that miR-8-positive glia express Spitz, a transforming growth factor alpha-like ligand that triggers epidermal growth factor receptor activation to promote neuroepithelial proliferation and neuroblast formation. PMID: 24139822
  2. Study describes crystal structures of s-dEGFR bound to its growth factor ligand Spitz, showing how occupying one binding site in a receptor dimer impairs ligand binding to the second site in an asymmetric complex. PMID: 20723758
  3. Argos inhibits EGFR (DER) signalling without interacting directly with the receptor, but instead by sequestering the DER-activating ligand Spitz PMID: 15329724
  4. role for palmitoylation in restricting Spitz diffusion, allowing its local concentration to reach the threshold required for biological function PMID: 16459296
  5. Argos mutants are deficient in their ability to bind Spitz in vitro PMID: 16870613
  6. The size of the posterior compartment in Drosophila embryoes is due, at least in part, to cells surviving and growing according to Spi availability. PMID: 17055987
  7. Spitz from the retina regulates genes transcribed in the second mitotic wave, peripodial epithelium, glia and plasmatocytes of the eye imaginal disc. PMID: 17553483
  8. Germ cells signal somatic support cells via the Egf class ligand Spitz. PMID: 17629483
  9. 1.6-A resolution crystal structure of Argos bound to Spitz PMID: 18500331
  10. interacts with Star and its function controlled by Star in the developing eye PMID: 11520660

Subcellular Location
Cell membrane; Single-pass type I membrane protein. Endoplasmic reticulum membrane; Single-pass type I membrane protein. Golgi apparatus membrane; Single-pass type I membrane protein. Note=Relocalization to the Golgi apparatus and the plasma membrane is mediated by Star. In the presence of Rhomboid, Spitz is found only in the Golgi, not at the cell surface.
Tissue Specificity
Expressed throughout the embryo.
