Target Background

Function
Required for meiotic recombination. Mediates DNA cleavage that forms the double-strand breaks (DSB) that initiate meiotic recombination. The action of SPO11 is important in setting off a regulatory chain of events encompassing 5' to 3' resection. When there are no SPO11-DSBs, resection of a site specific VDE-DSB takes place but it is faster than in wild-type meiosis and increases the risk of uncovering flanking homology.
Gene References into Functions
  1. Data show that increases in meiotic double-strand breaks and crossover frequencies can be programmed to arbitrary genomic sites by fusion of the Spo11 protein to various DNA-recognition domains. PMID: 28977556
  2. These findings provide an explanation for the unexpectedly low prototroph levels exhibited by spo11 hypomorphs and have important implications for genetic studies that assume an unbiased recovery of prototrophs PMID: 24204324
  3. Study shows that Spo11-accessory proteins Rec114, Mer2, and Mei4 stably interact with chromosome axis sequences, upon phosphorylation of Mer2 by S phase Cdk. PMID: 21816273
  4. meiotic double-stranded breaks in budding yeast are processed by endonucleolytic cleavage that releases Spo11 attached to an oligonucleotide with a free 3'-OH PMID: 16107854
  5. Analysis of mutation sites of isolated spo11-mutant alleles indicated that both N-terminal and C-terminal non-conserved residues of Spo11 are essential for the protein's function. PMID: 16816949
  6. Meiotic association between Spo11 regulated by Rec102, Rec104 and Rec114 occurs at the time of DNA double-stranded break formation. PMID: 17264124
  7. N-terminal part of Spo11p is a nuclear localization signal that is not specific for prophase I and is used to import proteins in vegetative yeast cells. PMID: 18702308
  8. How to detect these Spo11-oligo complexes in extracts made from meiotic yeast cells. PMID: 19799183
