Target Background

Function
Mitochondrial intermembrane chaperone that participates in the import and insertion of multi-pass transmembrane proteins into the mitochondrial inner membrane. Also required for the transfer of beta-barrel precursors from the TOM complex to the sorting and assembly machinery (SAM complex) of the outer membrane. Acts as a chaperone-like protein that protects the hydrophobic precursors from aggregation and guide them through the mitochondrial intermembrane space. Compared to TIM9, it may function as a substrate sensor.
Gene References into Functions
  1. Tim10 is protected from degradation by the protease Yme1 by Tim9. PMID: 26182355
  2. formation of the hexamer is beneficial but not vital for TIM10 function, whilst the N-terminal substrate-binding region of Tim10 is essential in vivo PMID: 17618651
  3. Assembly of the mitochondrial TIM9-TIM10 complex: a multi-step reaction with novel intermediates. PMID: 18022191
  4. oxidative folding and mitochondrial import are kinetically competitive processes; efficiency of mitochondrial import of the small Tim proteins is controlled, at least partially in vitro, by the rate of oxidative folding PMID: 18076384
  5. The structure of the yeast Tim9-Tim10 hexameric assembly determined to 2.5 A is reported and mutational analysis was performed to evaluate the specific roles of Tim9 and Tim10. PMID: 19037098
  6. A region within the Tim9 and Tim10 precursors, consisting of only nine amino acid residues, functions as a signal for the engagement of substrate proteins with the Mia40 receptor. PMID: 19297525
