Target Background

Function
Central component of the TOM (translocase of outer membrane) receptor complex responsible for the recognition and translocation of cytosolically synthesized mitochondrial preproteins. Together with TOM20 and TOM70 functions as the transit peptide receptor at the surface of the mitochondrion outer membrane and facilitates the movement of preproteins into the TOM40 translocation pore. Docks TOM20 and TOM70 for interaction with the general TOM40 import pore (GIP) complex. May regulate the TOM machinery organization, stability and channel gating.

  1. Tom22 is regulated by the cytosolic kinases CK2 and PKA and by membranebound CK1. PMID: 24093680

Study shows that the cytosolic domain of Tom22 links the outer mitochondrial membrane and the sorting and assembly machinery into a supercomplex, facilitating precursor transfer on the intermembrane space side and reveals receptor-mediated coupling of import and export translocases as a means of precursor channeling. PMID: 23911324
  3. We also identified a core sequence of Tom22 that binds to Tim21. Furthermore, negatively charged amino-acid residues of Tom22 are important for binding to Tim21. Here we suggest a mechanism for the TOM-TIM interaction. PMID: 17099692
