Target Background

Function

Predominant proteasome-associated deubiquitinase which releases ubiquitin from the proteasome targeted ubiquitinated proteins. Ensures the regeneration of ubiquitin at the proteasome. Has proteasome-inhibitory activity and delays the degradation of ubiquitinated proteins to provide a time window allowing gradual deubiquitination of the substrate. Stabilizes the association of HUL5 with proteasomes and works in opposition to polyubiquitin elongation activity of HUL5.

Electron microscopy analyses reveal that ubiquitin-bound Ubp6 contacts the N ring and AAA+ ring of the ATPase hexamer and is in proximity to the deubiquitinase Rpn11 in the proteasome. PMID: 26301997
Results suggest that Ubp6 regulates both the nature and magnitude of proteasome activity. PMID: 17018280
Results demonstrate that Sem1p and Ubp6p maintain telomeric heterochromatin structure through modulation of histone covalent modifications and association of silencing factors independently of the proteolytic function of the proteasome. PMID: 19188254
