Target Background

Function
Receptor for members of the ephrin family. Receptor for major sperm proteins (MSPs), that functions as sperm-sensing checkpoint which inhibits oocyte meiotic maturation and ovulation when sperm are not available for fertilization. Specifically, functions to negatively regulates oocyte maturation and MAPK activation in the absence of MSPs. Required for the MSP-mediated increase in the basal sheath cell contraction rate in somatic cells. Phosphorylates phosphatase daf-18/PTEN which probably promotes daf-18 degradation. By inactivating daf-18, regulates positively insulin-like daf-2 signaling cascade. Involved in interactions between neuronal substrate cells and a migrating epithelial sheet in head epidermis morphogenesis. Also required for cell movements following gastrulation and during ventral closure of the epidermis. Might play a role in spermatheca morphogenesis. Involved in axon guidance of SDQL neurons during neurogenesis.
Gene References into Functions
  1. Overall locomotion speed increased with increasing nicotine concentration in all tested mutant strains except in the vab-1 mutants PMID: 26574927
  2. Analysis of EFN-1(VAB2) expression and tissue-specific requirements is consistent with a model in which VAB-1 acts in amphid neurons, interacting with EFN-1 expressed on surrounding cells. PMID: 23979582
  3. VAB-1/Eph RTK can stop axonal outgrowth by inhibiting filopodia formation at the growth cone by activating Arp2/3 through a VAB-1/NCK-1/WSP-1 complex and by inhibiting UNC-34/Ena activity PMID: 22383893
  4. The germline apoptosis function of VAB-1 impacts specific cell death pathways, and may act in parallel to extracellular signal-regulated kinase MAPK signaling. PMID: 22240896
  5. Bridge formation requires VAB-1 kinase-dependent extension of presumptive bridge cells over scaffold cells toward the ventral midline during body wall closure PMID: 22197242
  6. identified the VAB-1 Eph receptor protein-tyrosine kinase as a major sperm protein receptor PMID: 12533508
  7. Data show that SAX-3/Robo functions with the VAB-1 Eph receptor for gastrulation cleft closure and ventral epidermal enclosure. PMID: 16033794
  8. VAB-1 switches from a negative regulator into a redundant positive regulator of oocyte maturation upon binding to MSP. NMR-1 mediates this switch by controlling UNC-43 CaMKII activation at the oocyte cortex. PMID: 16267094
  9. VAB-1 Eph RTK is required for targeting or limiting axons and neuronal cells to specific regions, perhaps by transducing a repellent or stop cue. PMID: 16386725
  10. This study shows that mutations in ina-1 (alpha-integrin), as well as vab-1 (Eph receptor), and vab-2 (ephrin), display defects in embryonic muscle cell migration. PMID: 18452914
  11. Regulated endocytic trafficking of the VAB-1 MSP/Eph receptor contributes to the control of oocyte meiotic maturation. PMID: 18472420
  12. Hypoxia exerted its [adverse] effect on axon pathfinding at least in part through HIF-1-dependent regulation of the Eph receptor VAB-1. PMID: 18587389

Protein Families
Protein kinase superfamily, Tyr protein kinase family, Ephrin receptor subfamily
Tissue Specificity
During ventral enclosure of the epidermis, expression is seen in clusters of presumptive head neuronal cells and several cells in the tail region. Early larvae show expression in the nerve ring and ventral nerve cord. Strong expression is also seen in the
