Target Background

Function
Essential for proper morphogenesis of the vacuole. May exist as structural reinforcement on the surface of the vacuolar membrane and be required for maintenance against rupture by osmotic pressure.
Gene References into Functions
  1. the Vam7 PBR affects PI3P binding by the PX domain and in turn affects binding to SNAREs and HOPS to support efficient fusion. PMID: 27365394
  2. first detailed characterization of the backbone dynamics of a PX domain, that of Vam7p, in the presence and absence of its ligand; the structure appears to tumble more rapidly in solution upon binding PtdIns(3)P, revealing a conformational change PMID: 16877709
  3. affinities of the PX domain for PtdIns(3)P and other lipids reveal a remarkable synergy within the multivalent complex that stably anchors Vam7p at the vacuolar membrane PMID: 16984909
  4. Data show that the ternary synergy of phosphoinositides and 2 SNARE chaperone systems (SNARE Vam7p and Sec17p/Sec18p/ATP and HOPS) is required for rapid fusion. PMID: 19805279
Subcellular Location
Vacuole.
