Target Background

Function
Acts as a transcriptional repressor, inhibiting expression in vitro. Probably redox-responsive. The apo- but not holo-form binds to its own promoter as well as that of groEL2. Oxidized apo-form and nitrosylated holo-form also bind DNA. The apo-form has been shown to act as a protein disulfide reductase, but also not to act as a protein disulfide reductase.
Gene References into Functions
  1. NMR structural model of WhiB1 reveals that Wbl proteins are four-helix bundles with a core of three alpha-helices held together by a [4Fe-4S] cluster. The WhiB1 structure suggests that loss of the iron-sulfur cluster (by nitrosylation) permits positively charged residues in the C-terminal helix to engage in DNA binding, triggering a major reprogramming of gene expression that includes components of the ESX-1 secretion ... PMID: 29273788
  2. we were able to unambiguously identify WhiB1, an essential transcriptional repressor capable of auto-repression, as a substrate of the mycobacterial Clp protease. PMID: 24603869
  3. Analysis of the C-terminal region of WhiB1 revealed the crucial roles played by a predicted beta-turn and two conserved positively-charged motifs in facilitating DNA-binding, but not iron-sulfur cluster acquisition. PMID: 22792304
  4. Data suggest whiB1 (with [4Fe-4S] cluster) is activated in presence of nitric oxide allowing whiB1 to bind to DNA (at region overlapping core promoter elements). PMID: 20929442
