Target Background

Function

Cleaves proteins C-terminally to mono- and paired-basic residues. Involved in the shedding of a subset of GPI-anchored plasma membrane proteins from the cell surface, including itself, GAS1 and MSB2. May also play a role in the maturation of GPI-mannoproteins associated with the cell wall. Can process the alpha-mating factor precursor. Required for cell wall integrity.

Yps1 N-entrance loop plays a major role in regulating the shedding activity of the endopeptidase, most likely by limiting access to the active site, and its cleavage in native Yps1 is associated with its shedding. PMID: 25886139
YPS1 expression is induced during cell wall stress and remodeling in a PKC1-MPK1-dependent manner, indicating that Yps1p is a direct, and important, output of the cell wall integrity response PMID: 16087741
The activation mechanism of GPI-anchored Yps1p was investigated and some of its substrates were identified. PMID: 18573178
Saccharomyces cerevisiae mucin Msb2p is processed into secreted and cell-associated forms, by the aspartyl protease Yps1p. PMID: 18591427
