The human HE-4 ELISA kit (CSB-E12923h) is designed for the quantitative measurement of human HE-4 protein in serum. It quantitates human HE-4 with 3.9 pg/ml sensitivity and shows excellent specificity for human HE-4. It uses the bi-antibody sandwich enzyme immunoassay technique. This assay employs a biotin-conjugated HE-4 antibody that recognizes the analyte bound by the immobilized HE-4 antibody, forming an antibody-analyte-antibody complex. The immune complex is further detected by avidin-conjugated HRP. The TMB solution is added into the wells and turns blue and finally turns yellow after the addition of the stop solution. Solution color develops in proportion to the amount of HE-4 in the sample. The O.D. value is measured using a microplate reader at 450 nm and is used to determine the concentration of the human HE-4 in the sample.
HE-4, encoded by the WFDC2 gene, is a secretory protein highly expressed in the human epididymis. As a proteinase inhibitor with protective immunity, HE-4 participates in the innate immune defense of multiple epithelia and in epithelial host defense. Human HE-4 is an important clinical biomarker used for the detection of epithelial ovarian cancer (EOC) due to its significant overexpression in EOC compared to normal tissue. It shows higher specificity and diagnostic accuracy than other biomarkers such as CA125. Overexpression of HE-4 has been detected in many tumors and is linked to cancer progression.

Regarding your ELISA kit CSB-E12923h, could you please advise on the following:
1) Is the standard natural material or recombinant? And if recombinant, what kind of cell or living material is it used for expression?
2) Does the standard include additive agent such as antiseptic or preservative? If included, what is the composition of additive agent?
3) Material name and concentration of Stop Solution.
4) Do these kits include hazardous substance such as azide, thimerosal, 2-ME, etc?
