This rat cyclooxygenase-1 (COX-1) ELISA kit employs the quantitative sandwich enzyme immunoassay technique to measure the levels of rat COX-1 in different samples, including serum, plasma, or tissue homogenates. The enzyme-substrate chromogenic reaction is also used to amplify the signal and quantify the levels of the analyte through the intensity of the colored product. The color intensity positively correlates with the amount of COX-1 bound in the initial step.
COX-1 (PTGS1) is a constitutive enzyme expressed in the vascular endothelium, in platelets, and in many other cells in the body. It is responsible for the initial cyclooxygenase reaction, the target for NSAIDs, the conversion of arachidonic acid to PGG2, and the subsequent peroxidase reaction that converts PGG2 to PGH2. Several studies suggest that COX-1 also plays a role in neuroinflammation. COX-1 also mediates prostaglandin-dependent gastric protection. In gastric mucosa, COX-1 is involved in the synthesis of PGE2 and PGI2, which exert cytoprotective effects on several aspects of gastric function such as an increase of bicarbonate and mucus secretion, reduction of gastric acid and pepsin secretion, and maintaining adequate blood flow to the mucosa.
