Interleukin 10 (IL-10) is a potent anti-inflammatory cytokine that plays an important role in immune regulation and homeostasis. Regulatory T cells, macrophages, and B cells produce IL-10 primarily. IL-10 works to suppress inflammatory responses by blocking pro-inflammatory cytokine production and reducing antigen presentation. This immunosuppressive activity makes IL-10 necessary for preventing excessive immune activation. It also helps maintain tissue integrity during inflammatory processes. The Sheep Interleukin 10 ELISA Kit (CSB-E12817Sh) measures IL-10 quantitatively in sheep samples using a competitive assay principle. This kit works with serum, plasma, and tissue homogenates. The detection range spans 5 pg/mL to 80 pg/mL with a sensitivity of 5 pg/mL. The assay needs 50-100 μL sample volume and takes 1-5 hours to complete. Detection occurs at 450 nm wavelength, giving researchers a reliable tool for IL-10 quantification in Ovis aries specimens.

Review: I used the CSB ELISA IL10 kit to detect IL-10 in synovial fluid of sheep. I tried it on normal samples, and osteoarthritis samples. I used a multiskan, with automatic conversion of absorbance, after automatic analysis (four parameter logistic curve-fit). I did not have a multichannel pipet so my values are not perfectly repeatable. The kit is easy to use, and protocol is clear. Just keep in mind that you have to read the plates at 450nm and correct with a 620-nm read. It is actually mentioned in the “other supplies required” section, but it is not remind in the “assay procedure” section, “assay procedure summary” section or in the “calculation of result” section (so during the assay there is a risk to forget the 2nd read).
