Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Ribonuclease (RNase) III involved in RNA-mediated post-transcriptional gene silencing (PTGS). Involved in the processing of repeat-associated small interfering RNAs (ra-siRNAs, derived from heterochromatin and DNA repeats such as transposons) by cleaving small dsRNAs into 24 nucleotide ra-siRNAs. Plays a role in antiviral RNA silencing. Involved in the production of viral siRNAs derived from the cabbage leaf curl virus (CaLCuV) and tobacco rattle virus (TRV). Targeted by the viral silencing suppressor (VSR) protein 2b of the cucumber mosaic virus (CMV) that inactivates DCL3 function in RNA silencing. Acts redundantly with DICER-LIKE 1 (DCL1) to promote flowering via repression of FLOWERING LOCUS C (FLC). Does not seem to be involved in microRNAs (miRNAs) processing.
Gene References into Functions
  1. Data demonstrate that different nutrient statuses of a host plant affect the post-translational regulation of the dicing activity of DCL3 and DCL4. Phosphate deficiency inhibited DCL3, and the activity of DCL3 was directly activated by inorganic phosphate. PMID: 28069892
  2. This expression-dependent RdDM mechanism functions through RNAi degradation of transposable element mRNAs into small RNAs guided by the RNA-dependent RNA polymerase 6 (RDR6) protein and is therefore referred to as RDR6-RdDM.We identified a new mechanism of expression-dependent RdDM, which functions through DICER-LIKE3 (DCL3) but bypasses the requirement of both RNA polymerase IV and RDR6 (termed DCL3-RdDM). PMID: 27506905
  3. The authors identified Pol IV and RDR2-dependent RNAs (P4R2 RNAs) that accumulate in dcl3 mutants and are diced into 24 nt RNAs by DCL3 in vitro. PMID: 26430765
  4. CMA33/XCT modulates small RNA production through regulating the transcription of Dicer-Like Genes. PMID: 25770820
  5. DCL3 preferentially cleaves short dsRNAs with 5' phosphorylated adenosine or uridine and a 1 nt 3' overhang. PMID: 24214956
  6. The data suggest a mechanism whereby IBM1 regulates a series of protein-coding genes including DCL3 through the RdDM-dependent control of siRNA biogenesis. PMID: 22772985
  7. DNA methylation establishment and maintenance defects present in sr45-1 mutants are enhanced in dcl3-1 mutant background, suggesting a synergistic cooperation between SR45 and DICER-LIKE3 (DCL3) in the RdDM pathway. PMID: 22274613
  8. Data show that small interfering RNAs and larger RNA fragments corresponding miRNA target genes accumulate in wild-type plants but not in ago1, the RNA-dependent RNA polymerase mutants rdr2 and rdr6, or the Dicer-like mutants dcl1 and dcl3. PMID: 16798886
  9. DICER-LIKE 1 and DICER-LIKE 3 are involved in the generation of small RNAs; mutants exhibit a delay in flowering. PMID: 17579240
  10. DCL3 is required for reception of long-distance mRNA silencing. PMID: 17785412
