Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Protein kinase that acts as a blue light photoreceptor in a signal-transduction pathway for photo-induced movements. Phosphorylates BLUS1, a kinase involved in stomatal opening. Mediates calcium spiking of extra- and intracellular origins in response to blue light. Involved in hypocotyl phototropism. Contributes to the chloroplast accumulation in low blue light and mediates their translocation (avoidance response) at high fluence. Regulates stomata opening and photomorphogenesis response of leaf tissue. Not involved in hypocotyl elongation inhibition, anthocyanin accumulation or cotyledon opening.
Gene References into Functions
  1. phot2 mediates the stabilization and relocation of NPH3 to the plasma membrane to acclimate to high-intensity blue light. PMID: 29393576
  2. This study demonstrates the photoinduced oligomerization of Arabidopsis thaliana phototropin 2 LOV1 domain. PMID: 25415534
  3. Phosphorylation of Ser-761 and Ser-763 in the activation loop may be a common primary step for phot2-mediated responses. PMID: 21427282
  4. Atphot2 behaves in a similar manner when expressed transiently under 35S or its native promoter. The phot2 kinase domain but not blue-light-mediated autophosphorylation is required for the receptor translocation. PMID: 24821953
  5. Phot2 mediates the high-intensity blue light-induced cytosolic calcium increases in hypocotyls.Phot2 ss required for phototropism under high-intensity blue light. PMID: 23674105
  6. Phot2 is the primary photoreceptor involved in the rapid reorganization of cloroplast-actin filaments that allows chloroplasts to change direction rapidly and control the velocity of the avoidance movement according to the light's intensity and position. PMID: 23404888
  7. RPT2 functions as a supporter for phot2 signaling. PMID: 22739508
  8. PHOT2 mediates the avoidance response of chloroplasts to stronger blue light (>32 micromol m-2 s-1). PMID: 22782888
  9. The C-terminal region of phot2 is responsible for the correct association with the chloroplast outer membrane. PMID: 23161859
  10. A small part of the C-terminal domain of phototropin 2 is necessary not only for membrane association but also for the physiological activities that elicit phototropin-specific responses. PMID: 23012349
  11. The analyses of the multiple phot2 mutant alleles provided strong evidence for the importance of both LOV domains and the kinase activation loop of phot2 in phototropism. PMID: 23196397
  12. In response to blue light, PHOT2 promotes the development of cylindrical palisade cells along a predetermined axis in a tissue-autonomous manner. PMID: 21972260
  13. Light-activated LOV2 domain triggers conformational changes in the linker region to separate the LOV2 and kinase domains. PMID: 21222437
  14. The light, oxygen, or voltage domain (LOV2) of phot2 plays a major role in phototropism and leaf expansion. PMID: 17085510
  15. analysis of a major conformational change of the linker region in the photochemical reaction of phototropin PMID: 17618649
  16. results indicate that phot2 mediates the blue light-dependent nuclear positioning and the proper positioning of nuclei and chloroplasts PMID: 17652112
  17. kinase activity associated with localization to functional sites and triggering of light signal transduction PMID: 17662032
  18. Data show that the expression level of 5PTase13 was significantly enhanced in phot1 single or phot1 phot2 double mutants under blue light, and suppression of 5PTase13 expression rescued the elongated hypocotyls in the phot1 or phot1 phot2 mutants. PMID: 18252844
  19. The recombinant LOV1 domains of phototropin 1 and phototropin 2 have been crystallized; the crystal of the LOV1 domain of phototropin 2 belongs to space group P2(1) and diffracts X-rays to beyond 2.0 angstroms resolution. PMID: 18607090
  20. Phot2 mediates the chloroplast avoidance response and chloroplast dark positioning. The N-terminal moiety of phot2 is most likely responsible for chloroplast positioning activity. PMID: 18643969
  21. Phototropism of inflorescence stems and leaf petioles is also controlled by phototropins. phot2 works predominantly under high light conditions. PMID: 19689999
