The creation of the recombinant candida albicans Glycerol-3-phosphate dehydrogenase [NAD(+)] 2 (GPD2) refers to the expression of the vector construct in yeast cells. The vector construct is established by cloning the gene fragment that codes for the full-length candida albicans GPD2 protein (1-371aa) along with a C-terminal 6xHis-tag into a plasmid vector. The vector construct containing the desired gene is transfected into the yeast cells. The transfected yeast cells are selected with a special antibiotic and then cultured in a suitable condition. The recombinant candida albicans GPD2 protein is expressed in the yeast cells at the action of the induction reagents. After that, the yeast cells are harvested and lysed to release the cellular contents, including the recombinant GPD2 protein. The recombinant GPD2 protein is purified from the cell lysate and assessed the purity of up to 85% through SDS-PAGE. It appears on a band with a molecular weight of 45 kDa on the gel.
