Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Essential for the development of polarized epithelia, for cell polarity associated with asymmetric cell division of neuroblasts during development, and for oocyte polarity formation. Promotes the formation of actin-rich projections at the oocyte cortex and the posterior enrichment of par-1 which is required for oocyte polarization. Regulates the localization of axis-specifying morphogens such as stau and grk.; Has an essential role in control of cell proliferation and differentiation during development and could act as a tumor suppressor.; Has an accessory function in control of cell proliferation and differentiation during development.
Gene References into Functions
  1. Here we show that PTEN mutation impacts aPKC and Lgl protein levels also in Drosophila. Moreover, we demonstrate that PI3K activation is not sufficient to trigger tumourigenesis, while aPKC promotes hyperplastic growth of the neuroepithelium and a noticeable expansion of the type II neuroblasts. PMID: 29445734
  2. Novel Genes Interacting with Lgl, aPKC, and Crb Cell Polarity Genes in Epithelial Tissues, is reported. PMID: 28611255
  3. results strongly suggest that the phenotypes we found in Sco mutants are more closely associated with the lgl allele than ectopic Snail activity PMID: 29261681
  4. Lgl plasma membrane (PB) domain contains all the identified phosphorylation sites of aPKC and Aurora kinases, providing a molecular mechanism by which phosphorylations neutralize the positive charges on the PB domain to inhibit Lgl PM targeting. PMID: 26483556
  5. AurA and aPKC exert the spatiotemporal control of Lgl distribution to achieve unique cell polarity roles in distinct cell types. PMID: 25484294
  6. Aurora A and B kinases directly phosphorylate Lgl to promote its mitotic relocalization. PMID: 25484300
  7. Lgl is a critical downstream target of the Par6-aPKC cell polarity complex. Lgl1 forms two distinct complexes in vivo, Lgl1-NMIIA and Lgl1-Par6-aPKCzeta in different cellular compartments. PMID: 25482644
  8. Rate of phosphorylation between sites differs at multiple sites in the atypical lethal giant larvae protein, regulating localization behaviors at the cortex. PMID: 25000553
  9. scrib and lgl expression in the somatic lineage of the Drosophila testis, similar to what was previously shown for dlg, was indispensable for testis development and homeostasis, as depletion of these genes resulted in extensive testes defects. PMID: 23585349
  10. presynaptic LGL scaffold facilitates the assembly of active zone fusion sites to regulate synaptic vesicle cycling, and that the postsynaptic LGL scaffold modulates glutamate receptor composition and function. PMID: 23444371
  11. Data indicate that loss of Lethal giant larvae (Lgl) tumor suppressor in the wing primordium induced epithelial neoplasia in its Homothorax (Hth)-expressing proximal domain. PMID: 23708122
  12. HSF1 and LGL interact during ontogenesis and stress-defending processes PMID: 22567869
  13. The p127 (l(2)gl)process nuclear proteins and then released from p127 (l(2)gl) by nmMHC to allow their binding to chromatin. PMID: 21989237
  14. The result reveals role for Lgl in neuronal specification and maintenance and show that the Hippo pathway is reimplemented for sensory neuron fate by combining canonical and noncanonical regulatory steps. PMID: 22055343
  15. These data indicate that Mahjong interacts with Lgl biochemically and genetically and that Mahjong and Lgl function in the same pathway to regulate cellular competitiveness. PMID: 20644714
  16. Data show that lgl mutant cells may form tumors if they acquire constitutive activity of the Ras pathway (lgl(-) UAS-ras(V12)), which confers proliferation advantage through inhibition of the Hippo pathway. PMID: 20679206
  17. Lgl depletion or aPKC overexpression results in comislocalization of Hippo and Ras-associated domain family protein (RASSF), consistent with RASSF's ability to block Hippo activation by Salvador. PMID: 20362447
  18. JNK signaling links lgl mutations to disruption of the cell polarity and epithelial organization in Drosophila imaginal discs. PMID: 20066009
  19. hitherto unreported role for Lgl as a regulator of Sanpodo during asymmetric cell division in the adult peripheral nervous system PMID: 15901829
  20. Lgl regulates cell fate by controlling Partner-of-Numb cortical localization, asymmetric localization of Numb and Neuralized, and plasma-membrane localization of Sandopo. PMID: 15916953
  21. Clonal analysis in larval brains showed that pins mutant neuroblasts rapidly fail to self-renew, whereas lethal giant larvae (lgl) mutant neuroblasts generate multiple neuroblasts PMID: 16357871
  22. The loss of one dose of tumor suppressor lgl+ provided for increased survival rate and life span of lgl/+ heterozygotes under stress conditions. PMID: 17352292

  25. Lgl and its phosphorylation by aPKC may form a conserved regulatory circuitry in polarization of various cell types. PMID: 18094021
  26. lgl is implicated in the formation of the initial AP asymmetry and the patterning of the AP and DV axes in the oocyte by acting in the specification of a subset of somatic follicle cells. PMID: 18268543
  27. Results show that embryonic exposure to heat stress and dosage of Lgl provide an adaptive advantage in later life in Drosophila. PMID: 18536270
  28. effect of impulse heating at the preembryonal stage was bidirectional: positive in the case of impulse warming of mature eggs and negative during egg differentiation. Deletion of one allele of the tumor suppressor gene in lgl/+ females increased survival PMID: 19239111
