What affinity tag was used at which site? Does the difference of tag location affect activity of the protein?
The tag type will be determined during production process. We can try to express your desired tag first if you have special request for the tag, but as we can't guarantee, if it's failed, we will commumnicate with you and provide the protein with other tag.
The difference of tag location may affect activity of the protein, but the probability is small.
Could you express EF4 with an N-terminal hexa-histidine (His6) tag? For the formulation buffer, could you try 10mM Citric Acid 6% Sucrose in pH 6.0?
Yes, we can try your desired N-terminal 6xHis-tagged first.
For buffer, we can try 10mM Citric Acid in pH 6.0. Instead of 6% Sucrose, we normally add 6% Trehalose as protective agent.
But there is no guarantee that the protein will be stable under this buffer condition. If it is unstable, we will communicate with you in adavance and provide default Tris/PBS/HEPES based buffer.
