Description

To prepare this Recombinant E.coli K12 ppk protein, the recombinant DNA was required, which was generated by fusing the ppk gene with N-terminal 6xHis tag sequence. Once the recombinant DNA was amplified and purified, a protein expression system, E.coli, was needed for this ppk protein production. After purification, a premium ppk recombinant proteinwas obtained. According to SDS-PAGE, its purity turns out to be 85%+.

ppk is a gene providing an instruction in making a protein named polyphosphate kinase. The encoded protein, also known as ATP-polyphosphate phosphotransferase or polyphosphoric acid kinase, is an enzyme that catalyzes the reversible transfer of phosphate molecules between ATP and polyphosphate (poly (P)). Emerging studies have found that the PPK catalyzes the kination of not only ADP but also other nucleoside diphosphates (NDPs) using poly(P) as a phosphate donor, yielding nucleotide triphosphates (NTPs). This protein is involved in three biological processes, including ATP generation from ADP, polyphosphate biosynthetic process and protein autophosphorylation.

Target Background

Function
Catalyzes the reversible transfer of the terminal phosphate of ATP to form a long-chain polyphosphate (polyP). Can form linear polymers of orthophosphate with chain lengths up to 1000 or more. Can use GTP instead of ATP, but the efficiency of GTP is 5% that of ATP. Also exhibits several other enzymatic activities, which include: ATP synthesis from polyP in the presence of excess ADP, general nucleoside-diphosphate kinase activity, linear guanosine 5'-tetraphosphate (ppppG) synthesis and autophosphorylation.
