Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

 Q&A
Q:

CSB-EP839870HU represents a serine protease. So if the protein is delivered in an active state, at some point it will probably digest itself and then is of no use for us anymore. There are ways however, to make sure it is not active (mutation of the active site, use of inhibitors to name a few). I know there are not many publications about PRSS33 so it is difficult to make predictions. But this is frankly speaking not our problem. So I want to be sure the protein is delivered in a good state and not degraded. Before we commit to an order, could you please check again how you will ensure the protein does not digest itself?

A:
Thank you for your e-mail. Here is reply from our tech team.
Indeed, for some a serine protease, it will digest itself. But it is influenced by many factors, such as protein expression and purification. We have successfully expressed two proteins from same protein family. CSB-EP859566MO(Prss29) and CSB-EP018817(Prss23). The purity is 85% and 90%, which shows the stability of such proteins. According to the previous experience in expressing the protein CSB-EP839870HU, the possible tag will be N-terminal 6×His-SUMO-tagged, we will optimize the expression and purification condition, and provide the soluble protein in stability form. Otherwise, the protein won't pass QC on our side.
On the other hand, we also refer to this protein on the market, we find that only GST tag is somehow degraded there is no self-degradation of the protein.

Refer to listed above points, there will be no big problem with the stability of the protein.
Q:

Human PRSS33 expressed in mammalian cells. What cell in particular were the host ones?

A:
Thanks for your inquiry. We usually choose the HEK293T cell to express the protein.

Target Background

Function
Serine protease that has amidolytic activity, cleaving its substrates before Arg residues.
Gene References into Functions
  1. activated eosinophils may induce fibroblast extracellular matrix protein synthesis through cell-surface expression of PRSS33, which would at least partly explain eosinophils' role(s) in airway remodeling. PMID: 28216055
  2. highly expressed in spleen and moderately expressed in intestine, colon, lung and brain PMID: 12795636
Subcellular Location
Secreted.
Protein Families
Peptidase S1 family
Tissue Specificity
Predominantly expressed in macrophages. Present in the spleen, small and large intestine, lung and brain (at protein level). Highly expressed in peripheral leukocytes, ovary, retina, spleen and stomach. Moderately expressed in thymus, uterus and platelets
