Reconstitution

We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage Condition

Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.

Description

Expressing the recombinant Kluyveromyces marxianus (Yeast) RPB5 protein in e.coli cells involves inserting a DNA fragment encoding the Kluyveromyces marxianus (Yeast) RPB5 protein (1-215aa) into a plasmid vector and transferring it to the e.coli cells. Positive cells are screened, cultured, and induced to express the RPB5 protein. The protein carries a N-terminal 6xHis tag. The cells are lysed to collect the recombinant Kluyveromyces marxianus (Yeast) RPB5 protein, which is purified through affinity purification and then identified using SDS-PAGE and subsequent staining of the gel with Coomassie Brilliant Blue. The purity of the resulting recombinant Kluyveromyces marxianus (Yeast) RPB5 protein exceeds 85%.

Function

DNA-dependent RNA polymerase catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates. Common component of RNA polymerases I, II and III which synthesize ribosomal RNA precursors, mRNA precursors and many functional non-coding RNAs, and small RNAs, such as 5S rRNA and tRNAs, respectively. Pol II is the central component of the basal RNA polymerase II transcription machinery. Pols are composed of mobile elements that move relative to each other. In Pol II, RPB5 is part of the lower jaw surrounding the central large cleft and thought to grab the incoming DNA template. Seems to be the major component in this process.
