Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Plays an early essential role in mesoderm formation, as well as a later role in formation of somite-derived chondrogenic lineages.
Gene References into Functions
  1. These results highlighted the functional importance of both transient and persistent expression domains of Scx for proper integration of the musculoskeletal components. PMID: 28327634
  2. Deletion of Scx led to altered long bone properties and callus healing. PMID: 27864378
  3. scleraxis was required for TGFbeta-induced fibronectin expression: TGFbeta lost its ability to induce fibronectin expression following scleraxis knockdown. PMID: 27324126
  4. The phosphorylation sequence matched that targeted by Casein Kinase 2, and inhibition of this kinase activity disrupted the ability of scleraxis to modulate the expression of its target genes while also attenuating TGFbeta-induced expression of type I collagen and myofibroblast phenotype conversion marker genes PMID: 26883788
  5. Cardiac fibroblast phenotype regulation by scleraxis. PMID: 26988708
  6. deletion of Scx led to impairments in enthesis structure. PMID: 26443819
  7. several mRNAs are affected by alternative splicing events in the absence of Scx, suggesting additional roles in post-transcriptional modification. PMID: 24983472
  8. Expression and function of scleraxis in the developing auditory system. PMID: 24058692
  9. These studies identify an important role for Scx in regulating proteoglycans in embryonic and mature valve cells. PMID: 24157418
  10. paratenon cells, which normally do not express Scx, respond to injury by turning on Scx and assembling matrix to bridge the defect PMID: 23555841
  11. Findings suggest a modular model for bone development, involving a distinct pool of Sox9- and Scx-positive progenitor cells that form bone eminences under regulation of TGFbeta and BMP4 signaling PMID: 23720048
  12. the Scx(+)/Sox9(+) progenitor pool is a unique multipotent cell population that gives rise to tenocytes, ligamentocytes and chondrocytes for the establishment of the chondro-tendinous/ligamentous junction PMID: 23615282
  13. Two proepicardial markers (Scleraxis and Semaphorin3D) were identified that genetically delineate heretofore uncharacterized proepicardial subcompartments. PMID: 22421048
  14. Mechanical signaling exerts an important influence on the expression of genes, namely scleraxis (Scx) and type I collagen, which play a role in determining the tendon phenotype. PMID: 21625049
  15. In addition to regulating the embryonic formation of limb tendons, scleraxis also appears to play an important role in the adaptation of adult tendons to physiological loading. PMID: 21913219
  16. Transforming growth factor-beta induces skeletal muscle atrophy and fibrosis through the induction of atrogin-1 and scleraxis. PMID: 22190307
  17. Scleraxis is required for differentiation of the stapedius and tensor tympani tendons of the middle ear. PMID: 21399989
  18. role in coordinating the response to injury in the pathogenesis of tendon disorders PMID: 20740671
  19. Murine fibroblasts expressing Scx mRNA from an exogenous Scx mini-gene indicated that the accumulation of Bop1 intronic RNA impairs the Scx gene expression in a trans-acting manner, which resulted in the reduction of the Scx mRNA level. PMID: 19996321
  20. Scx and E47 might modulate the primary chondrogenesis by associating with the Sox9-related transcriptional complex, and by binding to the conserved E-box on Col2a1 promoter. PMID: 19828133
  21. Study shows that tendon attachment to the skeleton regulates bone ridge formation and identify SCX regulation of Bmp4 as the underlying molecular mechanism. PMID: 20059955

  23. The phenotype of Scx-/- mutants emphasizes the diversity of tendon tissues and represents the first molecular insight into the important process of tendon differentiation. PMID: 17567668
  24. Scleraxis is required for cell lineage differentiation and extracellular matrix remodeling during murine heart valve formation in vivo. PMID: 18802027
