Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Applications : Drug related studies

Review: Canstatin suppresses hypoxia-induced apoptosis in H9c2 cardiomyoblasts through the activation of αvβ3 and/or αvβ5 integrins. H9c2 cardiomyoblasts were stimulated with hypoxia for 48 h in the presence or absence of canstatin (10±250 ng/ml) and cilengitide (1 μM), an αvβ3 and/or αvβ5 integrin inhibitor.

Applications : In vitro bioactivity in cell culture

Review: After the stimulation with isoproterenol (100 lM) for 48 h in the presence or absence of canstatin (250 ng/ml, 30 min pretreatment), differentiated H9c2 cardiomyoblasts were treated with 100 nM Mito Tracker Red for 30 min to observe mitochondrial morphology and membrane potential.

Applications : Drug related studies

Review: Acute effects of recombinant canstatin on the enhanced ICaL induced by COL4A2 siRNA in ventricular cardiomyocytes. Cardiomyocytes isolated from ventricles of COL4A2 siRNA-injected rats for 48 h were treated with canstatin (250 ng/ml) or solvent for 5-10 min.

Target Background

Function
Type IV collagen is the major structural component of glomerular basement membranes (GBM), forming a 'chicken-wire' meshwork together with laminins, proteoglycans and entactin/nidogen.; Canstatin, a cleavage product corresponding to the collagen alpha 2(IV) NC1 domain, possesses both anti-angiogenic and anti-tumor cell activity. It inhibits proliferation and migration of endothelial cells, reduces mitochondrial membrane potential, and induces apoptosis. Specifically induces Fas-dependent apoptosis and activates procaspase-8 and -9 activity. Ligand for alphavbeta3 and alphavbeta5 integrins.
Gene References into Functions
  1. alpha2(IV)NC1 domain has a role in regulating tumor cell behavior PMID: 15743801
  2. These results indicate that SOX9 is essential for Col4a2 expression in mesangial cells and might be involved in the accumulation of alpha2(IV) collagen in experimental nephritis. PMID: 17525254
Subcellular Location
Secreted, extracellular space, extracellular matrix, basement membrane.
