Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
May have an involvement in muscle development or hypertrophy. Isoform 2 binds to RBP-J and plays a negative regulatory role in the RBP-J-mediated transcription in mammalian systems.
Gene References into Functions
  1. KyoT2 downregulates airway remodeling and resistance in asthmatic mice through a Hes1-dependent mechanism. PMID: 26823730
  2. Results found that osteogenesis increases with the overexpression of Fhl1 and decreases with reduced Fhl1 expression and suggest that Fhl1 mediates interacting estrogen and Wnt signaling during osteogenesis. PMID: 25676585
  3. Findings indicate that loss of function is responsible for the myopathy in the Fhl1 W122S knock-in mice. PMID: 25274776
  4. The CSL-KyoT2 corepressor complex is a negative regulator of Notch signaling. PMID: 24290140
  5. Data show that loss of FHL1 function leads to myopathy in vivo and suggest that loss of function of FHL1 may be one of the mechanisms underlying muscle dystrophy in patients with FHL1 mutations. PMID: 23975679
  6. Fhl1 transcriptional changes provide salutary effects on stressed myocytes in cardiomyopathy. PMID: 24509080
  7. Data suggest Fhl1 (four-and-a-half LIM domains protein 1) and FHL2 (four-and-a-half LIM domains protein 2) bind to and regulate activity of Prkd1 (protein kinase D 1) in cardiac/ventricular myocytes; knockdown of Fhl1 down-regulates Prkd1 activation. PMID: 24219103
  8. A novel mechanism involving four-and-a-half LIM domain protein-1 and extracellular signal-regulated kinase-2 regulates titin phosphorylation and mechanics. PMID: 22778266
  9. Wnt signaling induces muscle cell differentiation, at least partly, through Fhl1 activation. PMID: 22367176
  10. FHL1 is a novel regulator of myosin-binding protein C activity that may have a role in sarcomere assembly PMID: 16407297
  11. These results suggest that KyoT2 is a substrate of SUMO modification catalyzed by PIAS1, and that SUMOylation may modulate the transcriptional repression effect of KyoT2 on the Notch/RBP-J signaling pathway. PMID: 17509614
  12. significantly altered expression after short-term hypoxic exposure; identified as a novel protein regulated in various forms of pulmonary hypertension PMID: 18725486
  13. KyoT3 is an isoform of murine FHL1, which associates with the transcription factor RBP-J and represses the RBP-J-mediated transactivation PMID: 18760388
  14. four-and-a-half LIM domains 1 (FHL1) is part of a complex within the cardiomyocyte sarcomere that senses the biomechanical stress-induced responses important for cardiac hypertrophy. PMID: 19033658
  15. FHL1 appears to modulate muscle mass and strength enhancement. PMID: 19075112
  16. Data show that SLIMMER delayed Siva-1-dependent apoptosis in C2C12 myoblasts. PMID: 19643733

Subcellular Location
[Isoform 1]: Cytoplasm.; [Isoform 2]: Nucleus.
Tissue Specificity
Isoform 1 seems to be most abundant in each tissue and isoform 2 much less abundant. Isoform 1 is highly expressed in skeletal muscle and lung, and to a lesser extent in heart, brain and kidney. Isoform 2 was found in brain, lung kidney and genital organs
