Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Receptor for interleukin-12. This subunit is the signaling component coupling to the JAK2/STAT4 pathway. Promotes the proliferation of T-cells as well as NK cells. Induces the promotion of T-cells towards the Th1 phenotype by strongly enhancing IFN-gamma production. Can also activate STAT3.
Gene References into Functions
  1. in neonatal mice, IL-12 uses IL-12Rbeta2 to counter IL-13Ralpha1 expression in addition to promoting Th1 differentiation PMID: 23650613
  2. survival of secondary LVS challenge was not strictly dependent on IL-12Rbeta2 PMID: 23440500
  3. Data indicate that delayed induction of the IL-12Rbeta2 receptor component after STAT1 activation helped ensure that Treg cells do not readily complete STAT4-dependent Th1 cell development and lose their ability to suppress effector T cell proliferation. PMID: 22960221
  4. IL-12R-beta2 signaling is essential for Plasmodium berghei ANKA-induced experimental cerebral malaria development PMID: 22238458
  5. Data show that T(H)1 differentiation was impaired in Il2(-/-) T cells but was restored by IL-12Rbeta2 expression. PMID: 21516110
  6. Freshly isolated ex vivo T helper (Th)17 cells display restricted expression from Il12rb2 due to the presence of repressive chromatin remodeling. PMID: 21307296
  7. T-bet mediates STAT1-dependent processes of T(H)1 development, including the induction of IL-12Rbeta2. PMID: 12006974
  8. NO activates soluble guanylyl cyclase, leading to the up-regulation of cGMP, which selectively induces the expression of IL-12 receptor beta2. PMID: 12451176
  9. Contrary to the expectation that the absence of IL-12RB2 would protect from experimental autoimmune encephalitis, IL-12RB2-deficient mice develop earlier and more severe disease, with extensive demyelination and central nervous system inflammation. PMID: 12574388
  10. expression of GATA-3 in developing Th1 cells suppresses Th1 development through downregulation of Stat4 rather through downregulation of the IL-12Rbeta2 chain PMID: 12648458
  11. IL-12Rbeta2-deficient mice of a genetically resistant background are susceptible to Leishmania major infection and develop a parasite-specific Th2 immune response. PMID: 12706437
  12. Expression of the IL-12 beta 2 receptor is upregulated in both microglia and splenic macrophages. PMID: 14568135
  13. By the third week of infection we found decreased IL-12Rbeta2 expression by BABL/c splenocytes, corresponding to their inability to produce interferon-gamma in Brucella recall responses at this time as reported previously. PMID: 17184756
  14. Contribution of IL-12rb2 mediated feedback loop to Th1 cell differentiation. PMID: 17950290
  15. IL-12Rbeta2 regulates both the number and functional maturity of regulatory T cells by a novel mechanism for the regulation of autoimmune diseases via the IL-12 pathway. PMID: 18768841
  16. Studies established the concept that the IL-12Rbeta2 gene is a gatekeeper from cancer. PMID: 19720917
  17. A point mutation in IL-12rb2 of the C57BL/10ScCr mouse produces a truncated beta2 chain which can still form a heterodimeric IL-12R that allows phosphorylation of Janus kinase 2 but is incapable of mediating STAT4 phosphorylation. PMID: 11489994
