Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Plays a role in intermediary metabolism and energy production. It may tightly associate or interact with the pyruvate dehydrogenase complex.
Gene References into Functions
  1. IDH2 knock out mice exhibit a decreased susceptibility to lipopolysaccharide (LPS)-induced lung injury. Depletion of IDH2 attenuates LPS-induced inflammatory response. PMID: 29913148
  2. role in maintaining the spinal structure by affecting the homeostatic balance between osteoclasts and osteoblasts PMID: 29527922
  3. our findings highlight the role of IDH2 in skin melanogenesis in association with mitochondrial ROS and suggest unique therapeutic strategies for the prevention of skin pigmentation. PMID: 29660504
  4. Data (including data from studies using knockout mice) suggest that IDH2 plays role in modulating liver fatty acid metabolism in dietary fructose-induced NAFLD (non-alcoholic fatty liver disease). This study was conducted in female mice; previous studies suggest that women are more prone to develop metabolic syndromes in response to fructose-sweetened beverages. PMID: 29861476
  5. present study supports the central role of idh2 deficiency in inducing oxidative stress resulting from acrolein-induced disruption of mitochondrial redox status in the lung PMID: 29456784
  6. IDH2 deficiency leads to increased mitochondrial ROS levels, activation of NF-kappaB and apoptosis of IEC in DSS-induced colitis. PMID: 28554049
  7. present study, we provided a role for IDH2 in protection against UVB-induced skin damage and a new connection between IDH2 and DeltaNp63. PMID: 29391208
  8. The disruption of IDH2 attenuates age-associated hepatic steatosis by the activation of p38/cJun NH2-terminal kinase (JNK) and p53, presumably induced by the elevation of mitochondrial reactive oxygen species (ROS), which in turn resulted in the suppression of hepatic lipogenesis and inflammation via the upregulation of fibroblast growth factor 21 (FGF21) and the inhibition of NFkappaB signaling pathways. PMID: 28415887
  9. We propose that D2-HG promotes cardiac dysfunction by impairing alpha-ketoglutarate dehydrogenase and induces histone modifications in an ACL-dependent manner.Combining mathematical modeling and in vivo as well as ex vivo studies, we found that increased amounts of the oncometabolite d-2-hydroxyglutarate (D2-HG), produced by IDH2 mutant leukemic cells, cause contractile dysfunction in the heart PMID: 27582470
  10. Increased obesity resistance and insulin sensitivity in mice lacking the IDH2 gene has been documented. PMID: 27519270
  11. treatment with Mito-TEMPO, a mitochondrial-specific superoxide scavenger, recovered mitochondrial fission-fusion imbalance and blunted mitochondrial superoxide production, and reduced the IDH2 knockdown-induced decrease in MnSOD expression, eNOS phosphorylation and NO production in endothelial cells PMID: 26898144
  12. Idh2R140Q mice displayed significantly elevated 2HG levels. PMID: 27469509
  13. These data support a significant role for increased oxidative stress as a result of compromised mitochondrial antioxidant defenses in modulating life span in mice, and thus support the oxidative stress theory of aging. PMID: 28928092
  14. Findings demonstrate that IDH2 contributes to degeneration of the DA neuron in the neurotoxin model of Parkinson's Disease. PMID: 27142242
  15. IDH2 deficiency promotes mitochondrial dysfunction and cardiac hypertrophy in mice. PMID: 25557279
  16. IDH2 and NPM1 mutations synergize in the development and maintenance of acute myeloid leukemia stem-like cells. PMID: 25795706
  17. 7-Ketocholesterol inhibits isocitrate dehydrogenase 2 expression and impairs endothelial function via microRNA-144 leading to atherosclerosis. PMID: 24642088
  18. These data demonstrate the proto-oncogenic role of mutant IDH2. PMID: 24440599
  19. The tumor tissue of B16F10 cells transfected with IDH2 antisense cDNA exhibited induction of apoptosis and downregulation of angiogenesis markers. PMID: 25086359
  20. Data indicate that tumor tissues from idh2-/- ((knock-out) mice had significantly decreased HIF-1alpha expression, blunted mRNA expression of HIF-1alpha, VEGF, and the glucose transport protein Glut-1 was also observed. PMID: 24240089
  21. IDH2 mutants can cooperate with oncogenic Flt3 or Nras alleles to drive leukemia in mice by impairing the differentiation of cells of the myeloid lineage PMID: 24065765
  22. SIRT3 protein deacetylates isocitrate dehydrogenase 2 (IDH2) and regulates mitochondrial redox status PMID: 22416140
  23. Results suggest that mitochondrial NADP(+)-dependent isocitrate dehydrogenase plays a role in apoptosis induced by high glucose and may contribute to various pathologies associated with the long-term complications of diabetes. PMID: 15522197
