Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Transcriptional repressor involved in multiple aspects of cortical development, including neuronal migration, laminar and areal identity, and axonal projection. As transcriptional repressor of FEZF2, it blocks the formation of the corticospinal (CS) tract from layer 6 projection neurons, thereby restricting the origin of CS axons specifically to layer 5 neurons.
Gene References into Functions
  1. Study induced neuronal differentiation of mouse bone mesenchymal stem cells (bMSCs) into neuron-like cells in vitro. miR-122-5p was well complementary with the target site of Tbr1 mRNA3'UTR and could inhibit Tbr1 expression by binding to its 3'UTR. Expressions of Tbr1 mRNA and protein were decreased. Overexpression of miR-122-5p downregulated the expressions of Tbr1, NSE, and TAU. PMID: 28240720
  2. CASK T740A mutation attenuated the interaction between CASK and TBR1 in the brain. However, CASK T740A mice were generally healthy, without obvious defects in brain morphology. The most dramatic defect among the mutant mice was in extinction of associative memory, though acquisition was normal. PMID: 28234597
  3. Mice lacking Tbr1 have Ca2+ waves in the cortex at normal frequencies and with normal neurotransmitter dependencies. PMID: 26473411
  4. Our findings highlight a TBR1-regulated network of autism spectrum disorders genes in the developing neocortex that are relatively intolerant to loss-of-function mutations, indicating that these genes may play critical roles in normal cortical development. PMID: 27325115
  5. T-Brain-1 (TBR1), a causative gene in autism spectrum disorders (ASDs), encodes a brain-specific T-box transcription factor. PMID: 25600067
  6. trans-synaptic Zn mobilization by clioquinol, a Zn chelator and ionophore, rescues the social interaction deficits in Shank2-/- and Tbr1+/- mice PMID: 25981743
  7. Transgenic overexpression of Wnt7b decreased expression of Tbr1 in both progenitor cells and post-mitotic neurons. PMID: 24953933
  8. This study demonistrated that Tbr1 haploinsufficiency impairs amygdalar axonal projections and results in cognitive abnormality. PMID: 24441682
  9. Results show that the expression of Tbr1 in neural stem and progenitor cells prevents them from following an astrocyte fate during olfactory bulb development. PMID: 20807572
  10. TBR1 is a direct transcriptional repressor of Fezf2 and a negative regulator of CS tract formation that restricts the laminar origin of CS axons specifically to L5. PMID: 21285371
  11. TBR1 promotes the identity of corticothalamic neurons and represses subcerebral fates through reducing expression of Fezf2 and CTIP2. PMID: 21228164
  12. Tbr1 coordinately regulates regional and laminar identity of postmitotic cortical neurons. PMID: 20615956
  13. Af9/Mllt3 interferes with Tbr1 expression through epigenetic modification of histone H3K79 during development of the cerebral cortex PMID: 20348416
  14. While Tbr1 is expressed in postmitotic neurons, Tbr2 (which is also expressed during gastrulation) is also expressed in neural progenitors. PMID: 12128215
  15. We suggest that Tbr-1/CASK protein complex regulates expression of these downstream target genes and thus modulates neuronal activity and function. PMID: 15584924
  16. These findings delineate the transcription factor sequence Pax6 --> Tbr2 --> Tbr1 in the differentiation of radial glia --> intermediate progenitor cell --> postmitotic projection neuron. PMID: 15634788
  17. Here, we provide a tool to illuminate a unique subset of SP and deep CP neurons: expression of a Tbrain-1 (Tbr1)-driven transgene. PMID: 16188453
  18. Tbr1 gene is expressed in cells that interact with olfactory sensory neurons PMID: 17506891
  19. Taken together, our data illustrate that Ngn2, neuroD and Tbr transcription factors are involved in postnatal neurogenesis in the olfactory bulb. PMID: 19200230
