Target Background

Function
Transcriptional activator involved in T-cell lymphocyte differentiation. Necessary for the survival of CD4(+) CD8(+) immature thymocytes. Isoforms lacking the N-terminal CTNNB1 binding domain cannot fulfill this role. Binds to the T-lymphocyte-specific enhancer element (5'-WWCAAAG-3') found in the promoter of the CD3E gene. Represses expression of the T-cell receptor gamma gene in alpha-beta T-cell lineages. Inhibits the developmental program of IL17A effector gamma-delta T-cell subsets via regulating the transcription of T-cell lineage effector proteins. Required for the development of natural killer receptor-positive lymphoid tissue inducer T-cells. TLE1, TLE2, TLE3 and TLE4 repress transactivation mediated by TCF7 and CTNNB1. May also act as feedback transcriptional repressor of CTNNB1 and TCF7L2 target genes.
Gene References into Functions
  1. Naive CD8+ T Cell TCF7 expression is FOXO1 independent. PMID: 28973925
  2. the inhibition of beta-catenin's TCF-dependent transcriptional activity, independent of its protein expression level, retains the naive ground state pluripotency in mouse embryonic stem cells. PMID: 28577307
  3. The balance between CD4(+) cytotoxic T cell and follicular helper T(Tfh) differentiation heavily depends on the class of infecting virus and is jointly regulated by the Tfh-related transcription factors Bcl6 and Tcf7 (encoding TCF-1) and by the expression of the inhibitory receptors PD-1 and LAG3. PMID: 27806296
  4. data reveal that T cell factor 1 (Tcf1) long and short isoforms have distinct, yet complementary, functions and may represent an evolutionarily conserved means to ensure proper programming of CD8(+) and CD4(+) T cell responses to viral infection PMID: 28652395
  5. Tcf7 levels are lower in islets taken from diabetic mice. Knockdown of TCF7 in islets impairs the cytoprotective responsiveness to GIP and enhances the magnitude of apoptotic injury. PMID: 26642437
  6. TCF-1-deficient CD4+ CD8+ double positive thymocytes fail to undergo TCR alpha Valpha14-Jalpha18 rearrangement and produce significantly fewer Natural killer T cells. PMID: 25536344

  10. TCF-1 was essential for both the initiation of T(FH) differentiation and the effector function of differentiated T(FH) cells during acute viral infection. TCF-1 promoted Bcl-6 expression but repressed Blimp1 expression. PMID: 26214740
