Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol.
Note: If you have any special requirement for the glycerol content, please remark when you place the order.
If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

 Q&A
Q:

I have purchased your VP1 Recombinant Protein (cat#: CSB-MP323858PQB; Lot#: 03542) and I believes that the Glycerol in these proteins is interfering with their binding assay. I would like to buffer-exchange it to get rid of the Glycerol. Do you have any recommendations regarding the buffer to use in order to avoid problems with solubility? Perhaps use PBS? Lyophilized?

A:
Very nice to receive your inquiry. As to your question, pls check our reply as follows:
CSB-MP323858PQB Buffer: 10 mM Tris-HCl, 1 mM EDTA, pH 8.0, 10% glycerol
As you can see, actually the glycerol concentration of these two items are already below our default shipping buffer 50% glycerol.
According to your feedback, since you already received these two items, does you want to change the buffer by yourself or need to order again ?
If you want to order again, we recommend you to choose the lyophilized form, the buffer before lyophilization will not contain any glycerol.
If you want to change the buffer or remove the glycerol by ourself, you can choose to remove the glycerol by dialysis or change the PBS buffer.
In addition, probably you can inform your application or requirements in advance next time, so that we can provide and supply the products according to his demand.
Buffer: 10 mM Tris-HCl, 1 mM EDTA, pH 8.0, 10% glycerol
Q:

Thank you for the information provided below. I would like to know the extinction coefficient of CSB-MP323858PQB respectively? I will need to measure the concentration after the buffer exchange in case of loss.

A:
Very nice to receive your inquiry. CSB-MP323858PQB
Extinctin coefficients are in units of M-1 cm-1, at 280 nm measured in water.
Ext. coefficient 13410
Abs 0.1% (=1 g/l) 0.607
Q:

1) Do you have any information on the stability of these two proteins below in PBS? The customer changed the buffer to PBS (got rid of glycerol) and stored them at -20C; do you think it may damage the proteins?
2) The customer may want to re-order and they would like to order in lyophilized format. They would like to know which buffer do you use to resolubilize the proteins? If the stability is not good after reconstitution, could it be possible to make aliquots?
3) Is it possible that the customer can order these tow proteins lyophilized and if they are not soluble following lyophilization then you can provide them in PBS?

A:
Very nice to receive your inquiry.
1.Sorry, I'm afraid we can't provide any useful information, since we haven't validated the stability in PBS buffer, But if your customer has already changed the buffer to PBS (got rid of glycerol) and stored them at -20C, will your customer use them after a long period ?
in theory, if the protein is already stable in it, this operation doesn't have any problem, and they will also be stable when stored at -20C.
2.Regarding resolubilizing the proteins, the reconstitution method is the same for all proteins. Please use deionized sterile water to reconstitute proteins.
(Dissolve completely. The concentration could depend on customers' experiment demand.) It is better that concentration does not be lower than 0.1mg/ml. Proteins will be unstable if they're too diluted.
Generally, we recommend 0.1-1.0mg/ml concentration. If customer has special requirements for concentration, then they could dilute the protein according to their experiment demand.
If the stability is not good after reconstitution, could it be possible to make aliquots?
Yes, we can make aliquots according to your request, pls remark your requirement when placing the order.
3.Yes, we can provide these two proteins in lyophilized format, generally, we will do reconstitution test for all lyophilized proteins before the shipment, if we find they are not soluble following lyophilization, then we will provide them in liquid form. We can provide the protein with your desired buffer (PBS buffer), pls remark your requirement when placing the order.

Target Background

Function
Capsid protein self-assembles to form an icosahedral capsid with a T=1 symmetry, about 22 nm in diameter, and consisting of 60 copies of two size variants of the capsid proteins, VP1 and VP2, which differ by the presence of an N-terminal extension in the minor protein VP1. The capsid encapsulates the genomic ssDNA. Capsid proteins are responsible for the attachment to host cell receptors. This attachment induces virion internalization predominantly through clathrin-dependent endocytosis. Binding to the host receptors also induces capsid rearrangements leading to surface exposure of VP1 N-terminus, specifically its phospholipase A2-like region and putative nuclear localization signal(s). VP1 N-terminus might serve as a lipolytic enzyme to breach the endosomal membrane during entry into host cell and might contribute to virus transport to the nucleus.
Gene References into Functions
  1. Data show the expressed VP2 caspid protein was secreted on the cell surface in Lactobacillus casei. PMID: 17460908
Subcellular Location
