Conjugate Avi-tag Biotinylated
E. coli biotin ligase (BirA) is highly specific in covalently attaching biotin to the 15 amino acid AviTag peptide. This recombinant protein was biotinylated in vivo by AviTag-BirA technology, which method is BriA catalyzes amide linkage between the biotin and the specific lysine of the AviTag.

Tag Info
Tag type will be determined during the manufacturing process.
The tag type will be determined during production process. If you have specified tag type, please tell us and we will develop the specified tag preferentially.
Form
Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer before Lyophilization
Tris/PBS-based buffer, 6% Trehalose, pH 8.0
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
Delivery time may differ from different purchasing way or location, please kindly consult your local distributors for specific delivery time.
Note: All of our proteins are default shipped with normal blue ice packs, if you request to ship with dry ice, please communicate with us in advance and extra fees will be charged.
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Target Background

Function
Ubiquitin-like modifier involved in the formation of autophagosomal vacuoles (autophagosomes). When lipidated mediates tethering between adjacent membranes and stimulates membrane fusion during autophagy. Recruits lipidated-lgg-2 to maturing autophagosomes. Acts in the aggrephagy pathway, which is the macroautophagic degradation of ubiquitinated protein aggregates, and preferentially interacts with autophagy proteins and substrates containing LIR motifs to mediate autophagosome formation and protein aggregate degradation. In particular, binds to components of the unc-51-atg-13 complex to regulate autophagosome formation and cargo sequestration. Required for the degradation of specific sepa-1- and sqst-1-containing protein aggregates during embryogenesis. Involved in allophagy, which is an autophagic process in which paternal mitochondria and organelles are degraded during fertilization, and moreover is required for the formation of lgg-2-positive allophagic autophagosomes in embryos. Involved in the clearance of apoptotic cells by promoting the delivery of engulfed apoptotic cells to the lysosome. Plays a role in the distribution and clearance of germ cell specific P-granules from somatic cells. Also plays a role in the autophagy-mediated degradation of ribosomal RNA and ribosomal proteins in lysosomes. Involved in xenophagy, the autophagy-mediated degradation of pathogens and pathogen products, such as toxins. Required for normal survival when exposed to pathogenic bacteria S.typhimurium probably by promoting autophagic degradation of intracellular S.typhimurium. Also plays a role in membrane-pore repair. Plays a role in mitophagy. Essential for dauer development and longevity, including longevity in response to moderate, short-term heat shock, also known as a hormetic heat shock.
Gene References into Functions
  1. Data show the N-terminal of LGG-1 and LGG-2 adopts the closed and open form, respectively. They have distinct binding pockets that prefer different residues, influencing binding specificity and differential interaction with autophagy proteins. PMID: 26687600
  2. During allophagy LGG-1 acts upstream of LGG-2 to allow its localization to autophagosomes. PMID: 24374177
  3. both LGG-2 and LGG-1 act synergistically and are involved in dauer formation and longevity of the worm. PMID: 20523114
  4. Inactivation of unc-51, bec-1 and lgg-1, the worm counterparts of the yeast autophagy genes Atg1, Atg6 and Atg8 respectively, partially suppresses degeneration of neurons with toxic ion channel variants. PMID: 17327275
Subcellular Location
Preautophagosomal structure. Cytoplasmic vesicle, autophagosome. Cytoplasmic vesicle, autophagosome membrane. Lysosome lumen. Mitochondrion. Cytoplasm. Cytoplasmic vesicle, phagosome membrane. Cell membrane; Lipid-anchor. Cell projection, dendrite. Perikaryon.
