Form
Liquid or Lyophilized powder
Note: We will preferentially ship the format that we have in stock, however, if you have any special requirement for the format, please remark your requirement when placing the order, we will prepare according to your demand.
Buffer
If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.
Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20℃/-80℃. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Troubleshooting and FAQs
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.
Shelf Life
The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself.
Generally, the shelf life of liquid form is 6 months at -20°C/-80°C. The shelf life of lyophilized form is 12 months at -20°C/-80°C.
Lead Time
3-7 business days
Notes
Repeated freezing and thawing is not recommended. Store working aliquots at 4°C for up to one week.

Description

The process of synthesizing the recombinant Pseudomonas aeruginosa (strain ATCC 15692 / DSM 22644 / CIP 104116 / JCM 14847 / LMG 12228 / 1C / PRS 101 / PAO1) eta protein includes transfecting e.coli cells with a DNA expression vector containing the gene for the desired protein (413-638aa). Subsequently, the cells are cultured to induce the expression of the intended protein. The protein is fused with a N-terminal 6xHis-KSI tag. The recombinant Pseudomonas aeruginosa (strain ATCC 15692 / DSM 22644 / CIP 104116 / JCM 14847 / LMG 12228 / 1C / PRS 101 / PAO1) eta protein is then collected and purified from the cell lysate through affinity purification. It exhibits a purity exceeding 85%, as verified by SDS-PAGE.

Target Background

Function
An NAD-dependent ADP-ribosyltransferase (ADPRT). Catalyzes the transfer of the ADP ribosyl moiety of oxidized NAD (NAD(+)) onto eukaryotic elongation factor 2 (eEF-2) thus arresting protein synthesis. Has an LD(50) of 65 ng/ml against the human lung epithelial cell line C38.
Gene References into Functions
  1. Our results show that the chimeric EGFETA toxin is extremely effective against EGFRpositive cancers and raises the potential to further develop this chimera for use in targeting EGFRpositive tumours resistant to monoclonal antibodies. PMID: 30226622
  2. Studied the presence of virulence genes (B-type flagellin [fliC], exotoxin A [toxA] and phzS) using molecular typing among isolates of Pseudomonas aeruginosa cultured from specimens from diabetic foot. Found fliC, toxA and phzS genes can play important roles in tissue penetration, tissue damage and survival. PMID: 29078729
  3. Data indicate that anti-EpCAM fusion toxin for tumor targeting by click PEGylation suggest new perspectives for effective cancer-related therapy. PMID: 24184633
  4. Use of the antibodies produced for testing toxigenicity of P. aeruginosa strains as well as detection of anatoxin in the technological process of production of prophylaxis preparations based on exotoxin A is expected. PMID: 24738291
  5. The highest level was observed for IL-1 and IL-6 after administration of recombinant proteins OprL, OprF, OprF-1, aTox. PMID: 24605680
  6. ATF-7 and ZIP-2 transcription factors are required for preventing ToxA-mediated lethality. PMID: 22520464
  7. These results indicated that exotoxin A acts as a virulence factor in Bombyx mori and that other virulence factor(s) are involved during the late stages of infection. PMID: 21945328
  8. When combined with IL-13Ralpha2 DNA cancer vaccine, Pseudomonas aeruginosa exotoxin A mediates synergistic antitumor effects on tumor growth and metastasis in established murine breast carcinoma and sarcoma tumor models. PMID: 22013118
  9. Norepinephrine represses the expression of toxA and the siderophore genes. PMID: 19686346
